Project description:Skeletal muscle possesses remarkable regenerative ability owing to the resident muscle stem cells (MuSCs). A prominent feature of quiescent MuSCs is a high content of heterochromatin. However, little is known about the mechanisms by which heterochromatin is maintained in MuSCs. We found that the mammalian Hairless (Hr) gene is expressed in quiescent MuSCs. Using a mouse model in which Hr can be specifically ablated in MuSCs, we demonstrate that Hr expression is critical for MuSC function and muscle regeneration. We show that Hr is a histone demethylase antagonist that preserves Histone 3 Lysine 9 trimethylation (H3K9me3) and maintains heterochromatin structure. Loss of Hr results in reduced H3K9me3 levels, reduced heterochromatin, increased susceptibility of MuSCs to genotoxic stress, and the accumulation of DNA damage. Our study not only elucidates the molecular mechanism by which Hr regulates histone demethylases, but also demonstrates the importance of heterochromatin maintenance in stem cell function.

Project description:The aging brain exhibits a decline in the regenerative populations of neural stem cells (NSCs), which may underlie age-associated defects in sensory and cognitive functions1-4 . While mechanisms that restore old NSC function have started to be identified5-9 , the role of lipids – especially complex lipids – in NSC aging remains largely unclear. Using lipidomic profiling by mass spectrometry, we identify age-related lipidomic signatures in young and old quiescent NSCs in vitro and in vivo. These analyses reveal drastic changes in several complex membrane lipid classes, including phospholipids and sphingolipids in old NSCs. Moreover, polyunsaturated fatty acids (PUFAs) strikingly increase across complex lipid classes in quiescent NSCs during aging. Lipidomic profiling of isolated plasma membrane vesicles shows that agerelated differences in complex lipid levels and side chain composition are largely occurring in plasma membrane lipids. Experimentally, we find that aging is accompanied by modifications in membrane biophysical properties, with a decrease in plasma membrane order in old quiescent NSCs in vitro and in vivo. To determine the functional role of plasma membrane lipids in aging NSCs, we perform genetic and supplementations studies. Knockout of Mboat2, which encodes a phospholipid acyltransferase, exacerbates age-related lipidomic changes in old quiescent NSCs and impedes their ability to activate. As Mboat2 expression declines with age, Mboat2 deficiency may drive NSC decline during aging. Interestingly, supplementation of plasma membrane lipids derived from young NSCs boosts the ability of old quiescent NSCs to activate. Our work could lead to lipid-based strategies for restoring the regenerative potential of NSCs in old individuals, which has important implications to counter brain decline during aging.

Project description:We performed single cell RNA sequencing on muscle satellite cells (MuSCs) and endothelial cells (ECs) isolated from mouse hind limb muscle from both basal condition and 3-days post CTX injury to look at both quiescent and activated SCs from mice expressing genetic reporters (Pax7CreERT2:R26RtdT:Flk1GFP) by FACS. We could reliably find injury and activated SC as judged by side and forward scatter. We FACS isolated cells from both days separately and spiked in 20% of the ECs into the SCs, and performed single cell RNA-seq for each time point. We performed extensive sequencing averaging more than 300K read/cell. We were able to align the data and overlap the basal and injured cells and able to deconvolve the quiescent SCs from the activated and more differentiated MuSCs, ECs and contaminating cell types from gene signatures.

Project description:Regeneration of skeletal muscle depends on a population of adult stem cells (satellite cells) that remain quiescent throughout life. Satellite cell regenerative functions decline with aging and in progeric conditions. Here we report that geriatric satellite cells, compared to old cells, are incapable of maintaining their normal quiescent state in muscle homeostatic conditions, and this irreversibly affects their intrinsic regenerative and self-renewal capacities. We analyzed the global changes in gene expression occurring within muscle stem cells (satellite cells) in homeostatic conditions during physiological aging and progeria. Pure satellite cell populations from dissociated skeletal muscle from Young (2-3 months), Old (22-24 months), Geriatric (28-30 months), SAMR1 and SAMP8 mice were isolated using a well-established flow cytometry protocol gating on integrin a7(+)/CD34(+) (positive selection) and Lin- (CD31, CD45, CD11b, Sca1) (negative selection).
