Deep RNA sequencing of HeLa secreted exosomes after 10ng/ml TGF-b1 treatment or no treatment

ABSTRACT: Deep RNA sequceing was performed to explore the expression level of miRNAs in HeLa cell exosomes after treatment with or without 10 ng/ml TGF-β1 for 24 h. The results showed that 48 miRNAs were enriched in exosomes of HeLa cells after treated with TGF-β1 compared with non-treated cells (negative control), proving that TGF-β1 affects HeLa cell exosomes miRNA profile.

TGF-β1 alter the SARS-CoV2 pathogenesis and modulates ACE2 expression by miRNA dependent mechanism

Project description:We studied miRNAs and their gene targets affecting SARS-CoV-2 pathogenesis in CF airway epithelial cell models in response to TGF-β1. Small RNAseq in CF human bronchial epithelial cell line treated with TGF-β1 and miRNA profiling characterized TGF-β1 effects on the SARS-CoV-2 pathogenesis pathways. Among the effectors, we identified and validated two miRNAs targeting ACE2 mRNA using different CF and non-CF human bronchial epithelial cell models. We have shown that TGF-β1 inhibits ACE2 expression by miR-136-3p and miR-369-5p. ACE2 levels were higher in cells expressing F508del-CFTR, compared to wild-type(WT)-CFTR and TGF-β1 inhibited ACE2 in both cell types. The ACE2 protein levels were still higher in CF, compared to non-CF cells after TGF-β1 treatment. TGF-β1 prevented the functional rescue of F508del-CFTR by ETI in primary human bronchial epithelial cells while ETI did not prevent the TGF-β1 inhibition of ACE2 protein. Finally, TGF-β1 reduced binding of ACE2 to the recombinant monomeric spike RBD. Our results may help to explain, at least in part, the role of TGF-β1 on the SARS-CoV-2 entry via ACE2 in the CF and non-CF airway.

Project description:TGF-β1 is involved in various biological processes through downstream signals including SMAD, Akt, Erk, etc. TGF-β1 overexpressed by harmful stimuli can act as a causative factor in diseases such as accumulation of extracellular matrix and development of cancerous characteristics. Therefore, we sought to compare changes in expression of intracellular miRNAs to study diseases caused by TGF-β1. In our study, we aimed to discover therapeutic targets for TGF-β1-induced disease by investigating changes in miRNA expression and identifying the correlation between genes that can be regulated by miRNA.

Global transcriptomic analyses of bronchial epithelial cells exposed to 5 ng/mL TGF-β1 and 10 nM Estrogen individually and in combination

Project description:To begin to identify downstream consequences of TGF-β1-induced reduction of estrogen receptor expression, we employed RNA-Seq. Results of global transcriptomic analysis showed that TGF-β1 and E2 inversely modulated the expression of several genes involved in both known pro-fibrotic cellular processes such as extracellular matrix turnover and newly identified events including airway smooth muscle cell contraction and calcium flux regulation. We also report, for the first time, that E2 specifically modulated the expression of genes involved in chromatin remodeling pathways and that this regulation was absent in the presence of TGF-β1. Collectively, these results suggest that E2 influences pathways relevant to pulmonary fibrosis and highlights potential roles for E2 in the lung that may contribute to sex-specific differences in IPF.

Gene expression profiling in acute murine autoimmune hepatitis

Project description:The etiology of autoimmune hepatitis is poorly understood but likely involves Th1 cells producing IFN-γ. BALB/c background TGF-β1-/- mice rapidly develop fulminant Th1-mediated autoimmune hepatitis. Our aims are to profile liver gene expression in TGF-β1-/- mice, to identify gene expression pathways dependent on IFN-γ as possible targets for rational therapy, and to test potential targets directly in vivo in mice. Keywords: Comparative analysis of gene expression in livers of WT, TGFB1 & IFN knockout mice DNA microarray analyses were applied to liver RNA from TGF-β1-/- mice, TGF-β1-/- /IFN-γ-/- mice, and TGF-β1+/+ littermate controls. 3 mice from each group were analyzed in this study.

Effects of LMTK2 knockdown on total RNA expression in CFBE41o- cells after TGFβ1 or vehicle control (HCl+BSA) treatment

Project description:The purpose of this study was to increase the current knowledge about the LMTK2 network and its intersection with the TGF-β1 signaling pathway. To understand how the LMTK2 and TGF-β1 pathways interconnect, we knocked down LMTK2 using small(si)RNA-mediated silencing in human bronchial epithelial CFBE41o- cells, treated cells with TGF-β1 or vehicle control, and performed differential gene expression analysis by RNA sequencing (RNAseq).
