Transcriptome analysis of striatal synaptosome and synaptosomal polyribosome of WT and Shank3 TG mice
Ontology highlight
ABSTRACT: Shank3 protein is a core organizer of the macromolecular complex in excitatory postsynapses, and its defects cause numerous synaptopathies including autism spectrum disorders. Although Shank3 function as a postsynaptic scaffold is well established, other potential mechanisms how Shank3 broadly modulates the postsynaptic proteome are relatively unexplored. To understand potential contribution of increased proteins synthesis to the proteomic change in TG striatal synaptosome, we performed RNA-seq analyses on both whole synaptosomal and synaptic polysome-enriched fractions. Comparative analyses showed a positive correlation only between polysome-associated transcriptome and up-regulated proteome in TG striatal synaptosome.
