RNA-seq analysis of intestinal organoids treated with SGC-CBP30 or vehicle at 36 hours post-irradiation

ABSTRACT: CBP/P300 inhibitors have been shown to be remarkably effective in mitigating radiation-induced GI syndrome both in ex vivo 3D organoid systemand animal studies. To further figure out the underlying mechanisms of improved radio-resistance by CBP/P300 inhibition, RNA sequencing (RNA-seq) was performed on crypt organoids treated with SGC-CBP30 or vehicle post 8Gy irradiation. RNA-seq analysis of crypt organoids demonstrated that a few radiation-activated pathways are significantly affected by SC treatment, such as E2F targets, G2M checkpoint, Myc targets, and DNA Repair. We speculate that the significant upregulation of genes in DNA repair pathway after CBP/P300 inhibition may help epithelial cells recovery from radiation damage.
