Project description:Aging changes global transcriptome including protein-coding and noncoding RNAs. Whether aging dependent lncRNAs undergo conserved regulation and function still remain unknown. We report aging dependent lncRNAs carry signature of evolutionary constraint and significantly enriched for targets of transcription factors and RNA binding proteins. Aging dependent lncRNAs is conservatively under NFκB pathway regulation. CRISPR screening for NFκB pathway found that aging dependent lncRNAs also extensively regulate NFκB pathway. The human LINC02085 is an evolutionary conserved lncRNA that can be traced to 29Mya before human. LINC02085 is induced during aging, inflammation and senescence through NFκB pathway. Reciprocally, LINC02085 directly regulates NFKBIZ in cis within same topological associated domain (TAD). LINC02085 binds to enhancer of NFKBIZ and recruits RELA to the NFKBIZ promoters and participates phase separation. These findings reveal that aging dependent lncRNAs are previously under-appreciated contributors to the evolutionary conservation and regulation of NFκB pathway.

Evolutionarily conserved odorant-binding proteins participate in establishing tritrophic interactions.

Project description:Attracting herbivores and their natural enemies is a standard method where plant volatiles mediate tritrophic interactions. However, it remains unknown whether the shared attraction has a shared chemosensory basis. Here we focus on the odorant-binding proteins (OBPs), a gene family integral to peripheral detection of odoriferous chemicals. Previous evidence suggests that the herbivorous beetle Monochamus alternatus and its parasitoid beetle Dastarcus helophoroides are attracted to stressed pines. In this study, (+)-fenchone, emitted by stressed pines, is found to be attracted to M. alternatus and D. helophoroides in behavioral assays. Meanwhile, two orthologous OBPs with a slower evolutionary rate, respectively, from the two insects are shown to bind with (+)-fenchone, and the attraction is abolished after RNAi. These results show the ability of evolutionarily conserved OBPs from herbivores and their enemies to detect the same plant volatiles, providing an olfactory mechanism of chemical signals-mediated tritrophic relationships.

Identification of evolutionarily conserved genetic regulators of cellular aging.

Project description:To identify new genetic regulators of cellular aging and senescence, we performed genome-wide comparative RNA profiling with selected human cellular model systems, reflecting replicative senescence, stress-induced premature senescence, and distinct other forms of cellular aging. Gene expression profiles were measured, analyzed, and entered into a newly generated database referred to as the GiSAO database. Bioinformatic analysis revealed a set of new candidate genes, conserved across the majority of the cellular aging models, which were so far not associated with cellular aging, and highlighted several new pathways that potentially play a role in cellular aging. Several candidate genes obtained through this analysis have been confirmed by functional experiments, thereby validating the experimental approach. The effect of genetic deletion on chronological lifespan in yeast was assessed for 93 genes where (i) functional homologues were found in the yeast genome and (ii) the deletion strain was viable. We identified several genes whose deletion led to significant changes of chronological lifespan in yeast, featuring both lifespan shortening and lifespan extension. In conclusion, an unbiased screen across species uncovered several so far unrecognized molecular pathways for cellular aging that are conserved in evolution.
