Transcriptomic characteristics underpin enhanced skeletal muscle mass, reduced fat accumulation and metabolically benign liver in human follistatin-344 transgenic pigs

ABSTRACT: Purpose: The goal of this study was to compare the mRNA transcriptomes of transgenic pigs and wild-type pigs for five organs (liver,longissimus dorsi,psoas major,subcutaneous fat and retroperitoneal fat). Methods: Total RNA was extracted from longissimus dorsi, psoas major, subcutaneous fat, retroperitoneal fat and liver using TRIZOL reagent (Life Technologies, Beijing, China), and each tissue has three biological replicates. For RNA sequencing, approximately 2 ug total RNA of each sample was used to construct transcriptome libraries following the manufacture’s standard procedures. Libraries were sequenced via Illumina HiSeqTM 4000, and 100 bp paired-end reads were generated. Results: We detected 18,051 protein coding mRNA transcripts across 30 samples .
