Gene regulation in the hyperoxia mouse retina

Project description:PURPOSE: Hyperoxia is toxic to photoreceptors, and this toxicity may be important in the progress of retinal dystrophies. This microarray study examines gene expression induced in the C57BL/6J mouse retina by hyperoxia over the 14-day period during which photoreceptors first resist, then succumb to, hyperoxia. METHODS: Young adult C57BL/6J mice were exposed to hyperoxia (75% oxygen) for up to 14 days. On day 0 (control), day 3, day 7, and day 14, retinal RNA was extracted and processed on Affymetrix GeneChip Mouse Genome 430 2.0 arrays. Microarray data were analyzed using GCOS Version 1.4 and GeneSpring Version 7.3.1. RESULTS: The overall numbers of hyperoxia-regulated genes increased monotonically with exposure. Within that increase, however, a distinctive temporal pattern was apparent. At 3 days exposure, there was prominent upregulation of genes associated with neuroprotection. By day 14, these early-responsive genes were downregulated, and genes related to cell death were strongly expressed. At day 7, the regulation of these genes was mixed, indicating a possible transition period from stability at day 3 to degeneration at day 14. CONCLUSIONS: Microarray analysis of the response of the retina to prolonged hyperoxia demonstrated a temporal pattern involving early neuroprotection and later cell death, and provided insight into the mechanisms involved in the two phases of response. As hyperoxia is a consistent feature of the late stages of photoreceptor degenerations, understanding the mechanisms of oxygen toxicity may be important therapeutically. 4 timepoints in total, 0d (control), 3d, 7d and 14d and a replicate per timepoint. TRKB Signaling Promotes Alveolar Capillary Angiogenesis Following Perinatal Hyperoxic Damage

Project description:Impaired angiogenesis characterized by the reduced proliferation of pulmonary endothelial cells leads to reduced capillary density in patients with bronchopulmonary dysplasia (BPD). In a mouse model of BPD, perinatal hyperoxic injury decreases the number of the recently identified lung capillary stem cells termed as general capillary (gCap) cells, along with the specific reduction of Ntrk2, which encodes for Tropomyosin receptor kinase B (TRKB) within this subpopulation. Herein, we determine whether TRKB signaling is required for perinatal gCap cell proliferation and further promoting pulmonary angiogenesis using the hyperoxia mouse BPD model. TRKB activation by BDNF treatment led to enhanced tube-forming ability of endothelial cells in vitro. In vivo treatment of mice with BDNF increased the proliferation of gCap cells and alleviated capillary loss caused by hyperoxic injury. Conversely, inhibition of TRKB signaling disrupted the tube formation of endothelial cells and exaggerated the vascular endothelial damage caused by hyperoxia. We further show that MAPK/ERK signaling might act downstream of TRKB to modulate pulmonary angiogenesis. These data indicate that TRKB signaling play a critical role in pulmonary angiogenesis upon perinatal lung injury, supporting the concept that TRKB activation might be a potential therapeutic for preserving endothelial cells for lung diseases associated with prematurity.

RNA-sequencing was carried out followed by differential expression of genes in the following groups. Heterogeneous mesenchymal cell types modulate neonatal lung development and its response to hyperoxia

Project description:During the first days of life, the lung surface area increases explosively to enable efficient oxygen exchange. Alveolarization and angiogenesis involve profound changes within the lung mesenchyme to modulate extracellular matrix composition, tissue elasticity, and coordination with the lung epithelium and endothelium. To define the cell types and states populating the perinatal lung mesenchyme, we performed single cell transcriptomics and in-situ imaging in mice and discovered an extremely heterogeneous and dynamic landscape of fibroblasts, airway smooth muscle-like, and mural cells spanning fourteen distinct populations. In the fibroblast and airway smooth muscle-like compartments, bipotent progenitors differentiate rapidly within one day of birth and then again towards the end of alveolarization, while mural cell types are characterized by slow, gradual changes. Glucocorticoid release, retinoic acid inactivation, and oxygen sensing are controlled by specific stromal progenitors. A population of myofibroblasts arises postnatally, peaks at the onset of alveolarization and disappears before three weeks of age. Paracrine signaling with dozens of lung cell types decreases after birth and especially during alveolarization. Nonetheless, alveolarization is the most proliferative developmental stage, followed by a shift towards cell quiescence. Exposure to hyperoxia during the first week of life delayed the transcriptomic changes of normal development, mirroring the arrested development observed in human neonatal bronchopulmonary dysplasia (BDP). Hyperoxia also caused a severe loss of pericytes and myofibroblasts and reduced their proliferation, decreased mesenchymal-endothelial communications, gave rise to a small novel population of contractile alveolar fibroblasts and increased matrix adhesion and contractility across the mesenchyme.
