ABSTRACT: ZKSCAN3 is an oncogenic transcription factor in a set of human malignant tumors including colon, prostate, bladder, and cervical cancers. We investigated that ZKSCAN3 acts as a downstream effector of oncogenic Wnt/beta-catenin signaling pathway. Deletion or knockdown of ZKSCAN3 increased MAD2L2 expression, resulting in delaying the cell cycle progression by increasing the time laps between prometaphase and telophase. Our study demonstrates that ZKSCAN3 mediates oncogenic Wnt/beta-catenin signals and induces chromosomal instability by suppressing MAD2L2 at the early stage of colon cancer development.
