Comparative analysis of gene expression after Oct4 or Scrambled siRNA knockdown in HNSCC cell lines

ABSTRACT: By employing a global gene expression profiling, we performed analysis of the molecular pathways which are deregulated in head-and-neck cancer cell lines FaDu, Cal33 and UTSCC5 after siRNA mediated knockdown of Oct4 comparing to Scrambled.

Project description:Mass spectrometry imaging (MSI) was used to investigate different tumors regions from five head and neck squamous cell carcinoma (HNSCC) xenograft samples of tumor model CAL33. Specifically, the authors investigated if measurements could be attributed to tumor hypoxia. Hypoxic regions were defined using consecutive tissue slices which were stained with anti-pimonidazole polyclonal antibody. M/z values which were associated to hypoxic regions were converted to masses and mapped with masses from LC-MS/MS experiment. Project description:To identify microRNAs induced by wogonin treatment, human head and neck squamos cell carcinoma cell lines were subjected to miRCURY LNA microRNA Array. FaDu and SAS cell lines were treated with 10 ug/ml wogonin or DMSO for 72h and total RNAs were extracted. Defferentially expressed microRNAs were analyzed by miRCURY LNA microRNA Array.

Project description:By employing a global gene expression profiling, we performed the differential gene expression analysis and the analysis of the molecular pathways which are deregulated in Hyperthermia (HT) and Radiation therapy (RT) treated SAS and FaDu spheroids from the head-and-neck cancer cell lines. The RNA-Seq analysis was done on FaDu and SAS spheroids treated with clinically relevant HT dose of 42.5°C for 60 minutes alone or in combination of single dose irradiation of 7 and 10 Gy for FaDu and SAS, respectively. The main pathways and networks modulated by the treatments (0.5 h after treatment) were identiﬁed using Gene Ontology (GO) enrichment analysis.
