Project description:Invariant NKT (iNKT) cells comprise a heterogeneous group of terminally differentiated, non-circulating, tissue-resident T-lymphocytes that recognize glycolipids, including alpha-galactosylceramide (aGalCer), in the context of CD1d. Here, we show that murine and human liver-resident aGalCer/CD1d-binding iNKTs correspond to a novel Zbtb16+/Tbx21+/Gata3+/Maflow/Rorc– subset that exhibits profound plasticity. Repetitive encounters of these cells with intravenously-delivered aGalCer/CD1d-coated nanoparticles (NPs) trigger their differentiation into immunoregulatory, IL-10/IL-21-producing, Mafhigh cells expressing a T-regulatory type 1 (TR1)-like transcriptional signature.

Project description:Unlike conventional CD4+ T cells, which are phenotypically and functionally plastic, invariant NKT (iNKT) cells generally exist in a terminally differentiated state. Although peripheral CD4+ T cells can acquire alternative epigenetic states in response to different cues, it remains unclear whether peripheral iNKT cells are epigenetically stable or maleable. Repetitive encounters of liver-resident iNKT cells (LiNKTs) with galactosylceramide (aGalCer)/CD1d-coated nanoparticles (NPs) can trigger their differentiation into a LiNKT cell subset (LiNKTR1) expressing a T regulatory type 1 (TR1)-like transcriptional signature. Here we use a broad range of omics tools to dissect the epigenetic underpinnings of the LiNKT-LiNKTR1 transdifferentiation process as compared to those underlying the T-conv-to-T-follicular helper (TFH) and TFH-to-TR1 differentiation pathways. We show that LiNKTR1 cell generation is partially associated with treatment induced hypo-methylation of gene bodies and regulatory elements of upregulated genes, in the absence of significant changes in chromatin exposure, deposition of expression-promoting histone marks, or removal of their repressive counterparts from bivalent regulatory elements. In contrast, the Tconv-TFH differentiation process involves extensive remodeling of the chromatin and the acquisition of a broad repertoire of epigenetic modifications that are then inherited by TFH cell-derived TR1 cell progeny. These results suggest that peripheral tissue-resident iNKT cells, unlike conventional T cells but like TFH cells, are epigenetically stable and that such stability contributes to their phenotypic and functional stability.

Project description:This study investigated the pulmonary toxicity of Copper oxide nanoparticles (CuO NPs) surface modified with polyethylenimine (PEI) or ascorbate (ASC), was investigated. Rats were exposed nose-only to a fixed exposure concentration of ASC or PEI coated CuO NPs for 5 consecutive days. On day 6 and day 27 post-exposure, pulmonary toxicity markers in bronchoalveolar lavage fluid (BALF) were analyzed and histopathological evaluation of the lungs was performed, along with microarray analyses on whole lung tissue samples.

Project description:Infections associated with antimicrobial-resistant bacteria now represent a significant threat to human health using conventional therapy, necessitating the development of alternate and more effective antibacterial compounds. Silver nanoparticles (Ag NPs) have been proposed as potential antimicrobial agents to combat infections. A complete understanding of their antimicrobial activity is required before these molecules can be used in therapy. Lysozyme coated Ag NPs were synthesized and characterized by TEMEDS, XRD, UV-vis, FTIR spectroscopy, zeta potential, and oxidative potential assay. Biochemical assays and deep level transcriptional analysis using RNA sequencing were used to decipher how Ag NPs exert their antibacterial action against multi-drug resistant Klebsiella pneumoniae MGH78578. RNAseq data revealed that Ag NPs induced a triclosan-like bactericidal mechanism responsible for the inhibition of the type II fatty acid biosynthesis. Additionally, released AgC generated oxidative stress both extra and intracellularly in K. pneumoniae. The data showed that triclosan-like activity and oxidative stress cumulatively underpinned the antibacterial activity of Ag NPs. This result was confirmed by the analysis of the bactericidal effect of Ag NPs against the isogenic K. pneumoniae MGH78578 1soxS mutant, which exhibits a compromised oxidative stress response compared to the wild type. Silver nanoparticles induce a triclosan like antibacterial action mechanism in multi-drug resistant K. pneumoniae. This study extends our understanding of anti-Klebsiella mechanisms associated with exposure to Ag NPs. This allowed us to model how bacteria might develop resistance against silver nanoparticles, should the latter be used in therapy.

Project description:ORganically MOdified SILica (ORMOSIL) nanoparticles (NPs) appear promising carriers for the delivery of drugs to target tissues and cells but some concerns on possible cytotoxic effects still exist. We therefore studied the in vitro responses to ORMOSIL NPs in different types of human lung cells (i.e. CCD-34Lu normal fibroblasts, carcinoma alveolar type II A549 cells, NCI-H2347 adenocarcinoma cells) to determine the effects of polyethylene glycol (PEG) coating on NP cytotoxicity. Our results show that non-PEG NPs caused a concentration-dependent decrease of viability of all types of cells. On the contrary, PEG-coated NPs increased plasma membrane permeability and induced cell death only in carcinoma alveolar type II A549 cells, while did not produce deleterious effects on CCD-34Lu and NCI-H2347 cells. PEG-coated NPs promoted the formation of reactive oxygen species (ROS) in A549 and CCD-34Lu cells; nevertheless, the decrease of ROS levels in the presence of superoxide dismutase and catalase did not protect A549 cells from death, suggesting that the oxidative stress was not the main determinant of cytotoxicity. Analysis of gene expression in A549 cells exposed to PEG-coated NPs showed alterations in genes involved in inflammation, signal transduction and cell death, while the transcriptional response was not significantly affected in CCD-34Lu fibroblasts. Our transmission electron microscopy analysis evidenced a unique intracellular localization of PEG NPs in the lamellar bodies of A549 cells, which could be the most relevant factor leading to cytotoxicity by reducing the production of surfactant proteins and by interfering with the pulmonary surfactant system.

Project description:To understand the the effect of poly(lactic-co-glycolic acid) nanoparticles (NPs) encapsulating a fluorine contrast agent on human umbilical vein endothelial cells (HUVECs), we have employed whole genome microarray expression profiling. The NPs used in this study were prepared in our lab. HUVECs were ordered from Lonza (Cat# C2517A). HUVECs were incubated with NPs for 4 hours. After 4 h, the cells were washed couple of times to remove the NPs and gene expression profiles were detected after 20 h (total time: 24 h) and 7 d in the absence of NPs. HUVECs after 24 h without nanoparticles were used as controls. The gene expression in HUVECs was measured at 24 h and 7 d after incubation with nanoparticles. Three independent experiments were performed at each time.

Project description:The current study deals to decipher the antibacterial mechanism of lysozyme coated silver nanoparticles (L-Ag NPs) (coated with lysozyme) against a Gram negative modal organism Escherichia coli K12 (MTCC 1302). Hence, the whole transcriptome profiling of E. coli K12 was done by exposing it to the MIC75 concentration of L-Ag NPs for 5 and 30 min., by RNA sequencing (RNAseq) analysis. The obtained results were utilized to understand all the metabolic pathways, signaling and molecular functions in bacterial cells under the stress of L-Ag NPs. RNAseq showed a high number of total reads along with significant ratio of high-quality reads, which confirmed the excellent quality and quantity of the obtained RNAseq data. Controlled release of ions from the surface of L-Ag NPs allowed the bacterial cells to function normally till the accumulation of threshold amount of silver ions which triggered the action of defence system, thus, reducing the chances of resistance development in bacteria. In long term, such treatment may force the bacterial machinery to induce changes in their genome to counteract the situation and develop resistance against silver ions, similar to the well-known antibiotic resistance problem. The obtained results advocate that L-Ag NPs can be used as effective antibacterial agent.

Project description:We used Au nanoparticles (Au-NPs) as a model for studying particle specific effects of manufactured nanomaterials (MNMs) by examining the toxicogenomic responses in a model soil organism, free living nematode Caenorhabditis elegans. Global genome expression for nematodes exposed to 4-nm citrate-coated Au-NPs at the LC10 (5.9 mg L-1) revealed significant differential expression of 797 genes. The levels of expression for five genes (apl-1, dyn-1, act-5, abu-11, and hsp-4) were confirmed independently with qRT-PCR. Seven common biological pathways associated with 38 of these genes were identified. Activation of 26 pqn/abu genes from noncanonical Unfolded Protein Response (UPR) pathway and up-regulation of molecular chaperones (hsp-16.1, hsp-70, hsp-3 and hsp-4) were observed and are likely indicative of endoplasmic reticulum stress. Inhibition of abu-11 with RNAi showed increase in mortality in Au-NP exposed nematodes suggesting possible involvement of abu-11 (a gene associated with specific to C. elegans UPR) in a protective mechanism against Au-NPs. Exposure to Au-NPs also caused activation of genes involved in apoptosis and necrosis and resulted ultimately in 10% mortality. These results demonstrate that Au-NPs are bioavailable and cause adverse effects to a model ecoreceptor which activate both general and specific biological pathways. Age-synchronized L3 nematodes (about 2000 nematodes per replicate with 3 replicates per treatment) were exposed to LC10 (5.9 mgL-1 ) of 4-nm citrate-coated Au-NP using 6 cm petri-dishes with 4 ml of exposure solution per dish. The N2 nematodes at L3 stageused were used and they were exposed in 50% K-Medium (31.68 mM KCl, 51.37 mM NaCl) for 12h. At 12 h all solutions were replaced with 50% K-Medium and left for another 12 h. After 24 h RNA was extracted from each of the replicates .
