Common features of aging fail to occur in Drosophila raised without a bacterial microbiome

ABSTRACT: By comparing the transcriptomes of Drosophila raised either under conventional conditions or in the complete absence of bacteria, we find that 70% of the transcriptional changes that normally occur with age are actually adaptive responses to the microbiome, not unavoidable features of aging. In the germ-free state, these seemingly ‘age-dependent’ gene expression changes never happen.

Project description:Lifespan is limited both by intrinsic decline in vigor with age and by accumulation of external insults. There exists a general picture of the deficits of aging, one that is reflected in a pattern of age-correlated changes in gene expression conserved across species. Here, however, by comparing gene expression profiling of Drosophila raised either conventionally, or free of bacteria, we show that ∼70% of these conserved, age-associated changes in gene expression fail to occur in germ-free flies. Among the processes that fail to show time-dependent change under germ-free conditions are two aging features that are observed across phylogeny, declining expression of stress response genes and increasing expression of innate immune genes. These comprise adaptive strategies the organism uses to respond to bacteria, rather than being inevitable components of age-dependent decline. Changes in other processes are independent of the microbiome and can serve as autonomous markers of aging of the individual.

Project description:A genome-wide association study was performed on ovaries from Siberian hamsters raised in either long or short photoperiod. Few differences between long and short photoperiod were noted at 3 wk of age, when ovarian histology was identical, whereas many differences in gene expression were noted at 8 wk of age, when ovarian histologies were markedly different. Hamsters were gestated and maintained in 16 or 10 hours of light per day, the latter short photoperiod delays sexual maturity and alters ovarian histology. Differences in gene expression were evaluated by microarray at 3 and 8 wk of age, as were changes between 3 and 8 wk of age in each photoperiod.

Project description:To better understand the hepatic metabolic response to intermittent fasting in chickens, Red Junglefowl chickens were raised on ad libitum (AL) feed until 14 days of age and then kept on AL feeding, switched to chronic feed restriction (CR) to around 70% or switched to an intermittent fasting (IF) regimen consisting of two fed days (150% of age-matched weight-specific AL intake offered daily) followed by a non-fed day. AL and CR were culled at 36 days of age, and IF birds either at 40 days of age (second consecutive feeding day) or 41 days of age (fasting day).
