Kras activation in endometrial organoids drives cellular transformation and epithelial-mesenchymal transition

ABSTRACT: We found that in KrasG12D-expressing endometrial organoids, Pten knockdown did not confer tumorigenicity, but Cdkn2a knockdown or Trp53 deletion led to the development of carcinosarcoma (CS), a rare, aggressive tumor comprising both carcinoma and sarcoma.To better characterize the pathogenesis of CS from organoids, we conducted a transcriptome analysis.

Establishment of novel murine cell lines with defined mutations for preclinical in vitro and in vivo investigations of syngeneic epithelial ovarian cancer with different histological subtypes and inflammatory profiles

Project description:Preclinical modeling of epithelial ovarian cancer in immune competent mice progressing to spontaneous tumors is challenging, requiring multiple genetic modifications in the host. We have generated a total of 28 immortalized cell lines with distinct genetic traits. Primary OSE cells were propagated in vitro and reached immortalization after an average of 8-10 months in culture. The cells were further engineered via deletion of Trp53, activation of oncogenic KrasG12D, deletion of Pten tumor suppressor or KrasG12D/Pten-/- combination. Cellular transformation was confirmed in vivo, via orthotopic tumor growth syngeneic hosts. Two different Trp53 null cell lines recapitulate high grade serous histology, Pten deletion triggers aggressive, high grade endometrioid tumors, and cells with dual KrasG12D activation and Pten deletion model carcinosarcoma. The cells express different tumor antigens, secrete various levels of chemoattracting cytokines and chemokines and trigger diverse in vivo inflammation profiles, including intratumoral T and B lymphocyte conglomerates. RNAseq data from 16 cell lines reveal the gene expression profile of several distinct models for high grade serous, endometrioid and carcinosarcoma histotypes. This versatile collection of murine cell lines significantly broadens the preclinical modeling capacity in ovarian cancer.

T-cell receptor repertoire in uterine and ovarian carcinosarcoma

Project description:Carcinosarcoma of the uterus or ovary is a rare, biphasic tumor comprising epithelial and mesenchymal elements, and exhibits aggressive clinical features. Four molecular subtypes of carcinosarcoma (POLE, MSI, CNH, and CNL) were recently established and shown to be associated with multiple clinicopathological parameters including patient outcomes. Immune microenvironment analyses on CS samples was performed using immune cell profiling with T-cell receptor repertoire assay. Carcinoma and sarcoma elements from CS samples were also assessed separately.

Mesenchymal tumor cells drive adaptive resistance of Trp53-/- breast tumor cells to inactivated mutant Kras

Project description:As precision medicine increases the response rate of treatment, tumors frequently bypass inhibition and reoccur. In order for treatment to be effective long term, the mechanisms enabling treatment adaptation need to be understood. Here, we report a mouse model that, in the absence of p53 and the presence of oncogenic KrasG12D, develops breast tumors. Upon inactivation of KrasG12D, tumors initially regress and enter remission. Subsequently, the majority of tumors adapt to the withdrawal of KrasG12D expression and return. KrasG12D-independent tumor cells show a strong mesenchymal profile with active MAPK/ERK signaling. Both KrasG12D-dependent and KrasG12D-independent tumors display a high level of genomic instability, and KrasG12D-independent tumors harbor numerous amplified genes that can activate the MAPK/ERK signaling pathway. Our study identifies both epithelial–mesenchymal transition (EMT) and active MAPK/ERK signaling in tumors that adapt to oncogenic KrasG12D withdrawal in a novel Trp53-/- breast cancer mouse model. To achieve long-lasting responses in the clinic to RAS-fueled cancer, treatment will need to focus in parallel on obstructing tumors from adapting to oncogene inhibition.

A Pancreas Organoids-based Screen Identifies a Specific KRAS Mutation Inhibitor that Attenuating Cholesterol Biosynthesis Pathway in Pancreatic Cancer

Project description:we reported a high-content drug screening platform of a series of pancreas organoids which contain common mutations found in human pancreatic cancer, wide type (WT), KC (KRASG12D), KPC (KRASG12D; Tp53R172H), KSC (KRASG12D; SMAD4-/-) and KPSC (KRASG12D; Tp53R172H and SMAD4-/-). Based on this platform, we screened about 6000 compounds and identified Perhexiline, which can inhibit the growth of pancreas organoids with KRAS mutation in vitro and in vivo. Single cell RNA-seq data shows that the cholesterol synthesis pathway is activated specifically caused by KRAS in mutated organoid lines. The key cholesterol synthesis regulator of SREBP2 and its downstream genes are activated in KRAS mutated organoids compared with WT. Interestingly, Perhexiline can reverse the metabolism changes in pancreas organoids with KRAS through downregulating SREBP2 at transcription level. Finally, we further validated targeting SREBP2 can attenuate tumor growth significantly with chemical inhibitor and genetic perturbation. These findings boost further understanding of KRAS in PDAC and provide a valuable resource for targeting KRAS therapeutics development.
