Project description:Brassinosteroids (BRs) are a group of plant steroid hormones that play crucial roles various of growth and developmental processes. Biological function and signal transduction pathway of BR has been well characterized in model plants like Arabidopsis and rice. However, their biological roles are still unclear in tree species, especially in an important non-timber plant moso bamboo, which has great ecological and economic value and distinguish fast-growth feature. Here we reported that reduce endogenous brassinosteroid by biosynthesis inhibitor propiconazole reduced both root and shoot growth in seedling stage. Exogenous BR application promoted shoot bract elongation and inclination of lamina joint and bract. Genome-wide transcriptome analysis were performed to identify hundreds of differential expressed genes by BR and propiconazole treatment in shoot and root parts of bamboo seedling, respectively. GO analysis revealed that BR regulates cell wall related genes, hydrogen peroxide catabolic genes and auxin related genes to promote bamboo root development and elongation. Our study identified BR response genes and provides a comprehensive resource for molecular mechanism research of bamboo growth.

Project description:Brassinosteroids (BRs) are a class of plant-specific steroid hormones which play key roles in plant physiology and seem to actively participate to the regulation of plant responses to stress. For this reason, these compounds are also grouped among the category of plant biostimulants which could be provided to crops to improve their performances and induce abiotic stress tolerance. In this study, thanks to a combined physiological and molecular approach new insight on the effects of 24-epibrassinolide (EBL), a synthetic BR, on hydroponically grown maize seedlings were gained. To this aim, a preliminary assessment of the effect of different EBL concentrations and time of treatment on root elongation was conducted to identify the concentration to be used in subsequent experiments. A treatment with EBL 1nM and a time exposure of 48 hours were then selected to better assess the effects of this molecule also on shoot growth and on root gravitropic response. Afterwards, a RNAseq based untargeted approach was applied to obtain an overview of the transcriptomic regulation occurring in roots, shoot and leaves upon exogenous brassinosteroids applications. Our outcomes highlight a deep influence exerted by this molecule on plant growth and on root gravitropic response of maize seedlings. Moreover, thanks to the functional characterization of differentially regulated genes by means of Gene Ontology (GO) and pathway analyses, new knowledge on the BR response and signalling in plants were gained. Globally, this study provides useful information which could help for future agriculture applications of these categories of substances.

Project description:The proper transition between the skotomorphogenic and photomorphogenic developmental programs is key for seedling survival and it is therefore tightly regulated. Besides light, which is the major cue that triggers this transition, several hormone pathways participate in this control, mainly antagonizing the light effect. Two of these hormones are gibberellins (GA) and brassinosteroids (BR). Both hormones promote the skotomorphogenesis and prevent photomorphogenesis, as evidenced by the partially de-etiolated phenotype caused by GA or BR deficiency, or by a blockage in the respective signaling pathways. We have previously shown that BR mediate GA activity in the control of this transition. For instance, treatment of dark-grown, GA-deficient seedlings with BRs was able to partially restore morphological phenotypes such as hypocotyl growth, and importantly to restore completely the molecular phenotype of CAB2 and RbcS gene expression. On the contrary, GA-treatment did not alter the same phenotypes in the BR deficient det2 mutant. Thus, the aim of this study was to investigate to what extent BRs mediate GA activity in dark-grown seedlings, and for that purpose we have the examined global changes in gene expression caused by treatment with GA and BRs in BR- and GA-deficient seedlings, respectively.

Project description:Brassinosteroids (BRs) are growth-promoting plant hormones that play a role in abiotic stress responses, but molecular modes that enable this activity remain largely unknown. Here we show that BRs participate in the regulation of freezing tolerance. BR signaling-defective mutants of Arabidopsis thaliana were hypersensitive to freezing before and after cold acclimation. The constitutive activation of BR signaling, in contrast, enhanced freezing resistance. Evidence is provided that the BR-controlled basic helix–loop–helix transcription factor CESTA (CES) can contribute to the constitutive expression of the C-REPEAT/DEHYDRATION-RESPONSIVE ELEMENT BINDING FACTOR (CBF) transcriptional regulators that control cold responsive (COR) gene expression. In addition, CBF-independent classes of BR-regulated COR genes are identified that are regulated in a BR- and CES-dependent manner during cold acclimation. A model is presented in which BRs govern different cold-responsive transcriptional cascades through the posttranslational modification of CES and redundantly acting factors. This contributes to the basal resistance against freezing stress, but also to the further improvement of this resistance through cold acclimation. We used microarray data to investigate the contribution of different pathways to cold tolerance of Arabidopsis thaliana .

Project description:Salt stress is a major environmental factor limiting plant growth and development. Previous studies have indicated that the steroidal hormones—brassinosteroids (BRs) are important regulators of plant responses to salt stress. We found that bes1-D, a gain-of-function mutant of the BR-regulated transcriptional factor BES1 (AT1G19350) in Arabidopsis showed better salt tolerance than the wild-type plant (Col-0). Label-free based LC-MS/MS and proteomic analyses were performed to investigate protein expression in bes1-D and wild-type in response to salt stress.

In this experiment, PAC samples were labelled with Cy3 and compared to WT and PAC+EBR samples, which were labelled with Cy5. Two biological repeast were performed. The second experiment included dark-grown, WT Col-0 seedlings and the det2-1 mutant, which was either mock-treated or treated with GA3. Two biological replicates were performed, in the first one the WT and det2-1+GA3 were labelled with Cy3 and the det2-1 sample with Cy5. The reverse labelling was used in the second replicate.

Project description:Plant hormone brassinosteroids (BRs) and abscisic acids (ABA) antagonistically regulate many aspects of plant growth and responses. This study analyzes the molecular mechanisms by which regulate the crosstalk between BR and ABA signaling.Various BR deficient and gain-of-function signaling mutants were used to analyze their responses to ABA inhibited primary root growth. RNA sequencing was performed to identify the ABA regulated root genes that are also regulated by BR signaling components.Our result demonstrated that BR signaling negative regulates plant ABA response, and the crosstalk is mediated by BIN2 and BZR1. RNA sequencing has identified subsets of ABA responsive root genes that were regulated by BIN2 and/or BZR1. ChIP-qPCR and EMSA assays showed that BZR1 could bind directly with several G-box cis-elements on ABI5 promoter, suppress the expression of ABI5 and makes plant insensitive to ABA.These data demonstrated that ABI5 is a BZR1 direct targeted gene. Regulation of ABI5 expression by BZR1 plays important roles in regulating the crosstalk between BR and ABA signaling pathways.

Project description:Plant photomorphogenesis and skotomorphogenesis depend on interplay of various plant hormones. Brassinosteroids (BRs) play crucial roles during hypocotyl establishment both in the light and darkness. Arabidopsis brassinosteroid insensitive1 (bri1-5) is a partial loss-of-function mutant, and the short-hypocotyl phenotype of bri1-5 is greatly suppressed by a long-hypocotyl mutant phytochrome B (pbyB-77) at the double mutant for the two in the light conditions to display additive phenotypes of both. To identify the genes responsible for this genetic interaction, we employed microarray analysis with the RNAs prepared from wild type, bri1-5, phyB-77, and bri1-5 phyB-77 double mutant. Microarray analysis supported the notion that PHYB acts upstream of BRs.
