Project description:In other bacteria, arginine induces the expression of genes involved in arginine catabolism. This study obtained the identification of genes involved in the arginine metabolism of Mycobacterium tuberculosis. Mycobacterium tuberculosis was cultured with arginine or ammonium chloride as sole nitrogen source. In the log phase of growth, RNA was isolated and whole genome expression was determined. The study contains three biological replicates.

Project description:Paraffin-embedded lung and spleen tissues analyzed by Eksigent nanoLC-Ultra 2D System and QExactive mass spectrometer. Both lung and spleen tissues were extracted from animals at 4 different conditions (Not infected Ad libitum, Not infected Caloric restricted, Mycobacterium Tuberculosis (MTB) infected Ad libitum, Mycobacterium Tuberculosis (MTB) infected Caloric restricted). Globally, 24 and 23 runs are uploaded for lung and spleen tissues, respectively.

An alveolus lung on a chip model of Mycobacterium fortuitum lung infection

Project description:Lung disease due to non-tuberculous mycobacteria (NTM) is rising in incidence. While both 2D cell culture and animal models exist for NTM infections, a major knowledge gap is the early responses of human alveolar and innate immune cells to NTM within the human lung microenvironment. Here we describe development of a humanized, 3D, alveolus lung-on-a-chip (ALoC) model of Mycobacterium fortuitum infection that incorporates only primary human cells such as pulmonary vascular endothelial cells in a vascular channel, and type I and II alveolar cells and monocyte-derived macrophages in an alveolar channel along an air-liquid interface. M. fortuitum introduced into the alveolar channel primarily infected macrophages, with rare bacteria inside alveolar cells. Bulk-RNA sequencing of infected chips revealed marked upregulation of transcripts for cytokines, chemokines and secreted protease inhibitors (SERPINs). Our results demonstrate how a humanized ALoC system can identify critical early immune and epithelial responses to M. fortuitum infection. We envision potential application of the ALoC to other NTM and for studies of new antibiotics

Histone arginine methylation by Prmt controls lung branching morphogenesis through transcriptional repression of Bmp

Inactivation of Prmt in the lung epithelium results in halted branching morphogenesis, altered P-D airway patterning and neonatal lethality.

Histone arginine methylation by Prmt5 is required for lung branching morphogenesis through repression of BMP signaling

Project description:Branching morphogenesis is essential for the successful development of a functional lung to accomplish its gas exchange function. Although many studies have highlighted requirements for the bone morphogenetic protein (BMP) signaling pathway during branching morphogenesis, little is known about how BMP signalingis regulated. Here we report that the protein arginine methyltransferase 5 (Prmt5) and symmetric dimethylation at histone H4 arginine 3 (H4R3sme2) directly associate with chromatin of Bmp4 to suppress its transcription. Inactivation of Prmt5 in the lung epithelium results in halted branching morphogenesis, altered epithelial cell differentiation and neonatal lethality. These defects are accompanied by increased apoptosis and reduced proliferation of lung epithelium, as a consequence of elevated canonical BMP-Smad1/5/9 signaling. Inhibition of BMP signaling by Noggin rescues the lung branching defects of Prmt5 mutant in vitro. Taken together, our results identify a novel mechanism through which Prmt5-mediated histone arginine methylation represses canonical BMP signaling to regulate lung branching morphogenesis.

Arginine to Cysteine substitutants in lung cancer enhance survival to chemotherapy

Project description:Tryptophan and arginine are the two most prominent restrictive amino acids linked to anti-tumor activity. For tryptophan, the induction of indoleamine 2,3-dioxygenase 1 (IDO1) by interferon-gamma (IFNg) secreted by T cells, catabolizes tryptophan to kynurenine to suppress T cell activity. For arginine, tumor cells suppress Argininosuccinate Synthase 1 (ASS1) expression to limit arginine availability for T cell activity (26560030). While tryptophan shortage induces tryptophan to phenylalanine (W>F) substitutants in the proteomes of a variety of tumor types (35264796), whether arginine shortage induces arginine substitutants is unknown. Here, we interrogated the proteomes of cancer patients for arginine substitutions and identified a strong enrichment for cysteine (R>C) specifically in lung tumors. Using a biochemical assay and mass spectrometry, we validated the induction of R>C substitutants by arginine shortage and indicated their link to intracellular high cysteine levels. In the context of lung cancer, R>C events did not overlap with R>C mutations, while a strong correlation with ferroptosis genes and with oncogenic mutations related to the Kelch-like ECH-associated protein 1 (KEAP1) ferroptosis pathway was observed. Indeed, KEAP1 mutations induce intracellular cysteine levels (10.7554/eLife.45572), and the expression of intact KEAP1 in KEAP1-mutated cells suppressed R>C substitutants. We further underpinned tRNA misalignment as the underlying mechanism for R>C events, and, show that boosting R>C serves to enhance resistance to chemotherapy. Thus, our work identified a novel mechanism of enriching proteomes with cysteines, which is exploited by arginine shortage in lung cancer to better endure chemotherapy stress.
