Single Transcriptional Factor (PU.1) Efficiently Leads Human iPS Cells to Functional Microglia

ABSTRACT: We developed an efficient method of generating microglia (hiMGLs) from human induced pluripotent stem cells (iPSCs) through forcing the expression of single transcription factor, PU.1. By using this method, we were abled to harvest a great deal of microglia in about three weeks. Transcriptome profile revealed that our hiMGLs well resembled characteristics of human primary microglia. Furthermore, we confirmed that transient over-expression of PU.1 would not change gene expression of microglia in central nervous system or neurological diseases.
