Project description:We investigated root associated fungi in young Norway spruce (Picea abies) cuttings rooted from slow- and fast-growing trees showing variable growth rate in long-term field experiments and compared their roots’ gene expression patterns five and 18 months after adventitious root initiation. Gene expression patterns of adventitious roots could not be systematically linked with the growth phenotype at the initiation of root formation, and thus fundamental differences in the receptiveness of fungal symbionts could not be assumed.

RNAseq evaluation of nicotinamide seed treatment analysed in Norway spruce seedlings

Project description:Nicotinamide seed treatment is investigated as a method for production of robust plants. Previous studies have shown that nicotinamide promotes the plant defence system and decreases DNA methylation levels. The purpose of the present work was to learn more about the underlying molecular mechanisms. The RNA was extracted from roots of three months old Norway spruce seedlings treated at seed stage. Stress was an overrepresented biological process of upregulated genes. Stress response genes are involved in adaption to and defence against stress. Upregulated stress response genes in the seedling roots were for example transcription factors MYB77 and LHY and two chitinases. Chitinases defend against biotic stress, such as pine weevils. Epigenetic regulating genes were generally downregulated, such as DDM1 which is known to promote DNA-methylation. Previous works suggest the hypothesis that nicotinamide is a defense signal mediator, which is supported by the present study.

Effect of flg22 peptide on Arabidopsis seedling roots with endodermis-expressed FLS2 in fls2 mutant background

Project description:To compare transcriptional outputs of FLS2 and SGN3 pathways solely within the endodermis, we conducted comparative RNAseq analysis of pCASP1::FLS2 fls2 and wild-type seedling roots treated by flg22 and CIF2 respectively, at three timepoints (30, 120, 480 min) (CIF2 responses data previously published by Fujita et al 2020). The receptor mutant controls sgn3 and fls2, displayed minimal gene responses to respective peptides.

Trehalose determination in Norway spruce (<i>Picea abies)</i> roots. Analytics matters.

Project description:We present concise results of method validation for trehalose quantitation by LC-MS/MS in spruce ectomycorrhizal roots in order to describe spruce health status, mainly in connection to contamination by a pathogenic fungus, Gemmamyces piceae. The procedure is based on Rogatsky et al. (2005) developed for human plasma. We found out that the best extraction yield was achieved with 80% methanol/water (v/v) solution and optimal extraction temperature was set between 50-60°C. In contrast to previous papers, we minimized the activity of trehalase enzyme by putting root samples into liquid N2 immediately after root excavation, followed by freeze-drying in order to stop trehalase activity. Higher content of trehalose was recorded in healthy trees, confirming the idea that ectomycorrhiza plays a significant role in plant-pathogen interactions.

Stability and Photoisomerization of Stilbenes Isolated from the Bark of Norway Spruce Roots.

Project description:Stilbenes or stilbenoids, major polyphenolic compounds of the bark of Norway spruce (Picea abies L. Karst), have potential future applications as drugs, preservatives and other functional ingredients due to their antioxidative, antibacterial and antifungal properties. Stilbenes are photosensitive and UV and fluorescent light induce trans to cis isomerisation via intramolecular cyclization. So far, the characterizations of possible new compounds derived from trans-stilbenes under UV light exposure have been mainly tentative based only on UV or MS spectra without utilizing more detailed structural spectroscopy techniques such as NMR. The objective of this work was to study the stability of biologically interesting and readily available stilbenes such as astringin and isorhapontin and their aglucones piceatannol and isorhapontigenin, which have not been studied previously. The effects of fluorescent and UV light and storage on the stability of trans stilbenes were assessed and the identification and characterisation of new compounds formed during our experiments were carried out by chromatographic (HPLC, GC) and spectroscopic techniques (UV, MS, NMR). The stilbenes undergo a trans to cis isomerisation under extended UV irradiation by intramolecular cyclisation (by the formation of a new C-C bond and the loss of two hydrogens) to phenanthrene structures. The characterised compounds are novel and not described previously.

Gene expression profiling of ramie roots during hydroponic induction and adaption to aquatic environment

Project description:Transcriptome analysis of root development related genes in terrestrial and hydroponic ramie. Ramie seedlings were cultivated in soil, and in hydroponic with the shoot-cutting propagation method. The root samples from hydrophonic ramie were collected from the early stage (5-day-old seedling) and the late stage (30-day-old seedling) of acquatic roots induction. The roots of ramie seedling cultivated in soil were also collected for comparative analysis. Our study represents the detailed analysis of ramie root transcriptomes with biologic replicates.
