Next generation sequencing facilitates quantitative analysis of transcriptional profiling of Candida albicans treated with thiosemicarbazone derivative 19ak.

ABSTRACT: Candida albicans SC5314 cells were treated with 19ak for mRNA profiling analysis through deep sequencing, in triplicate, using Illumina HiSeq X Ten. The sequence reads that passed quality filters were analyzed at the transcript isoform level. qRT–PCR validation was performed using SYBR Green assay. We find that genes invovled in mitochondrial respiration such as NUO1, NUO2 and MCI4 and ribosomal biogenesis such as RPL5,RPS43,RPS12 were downregulated. Genes invovled in reduction-oxidation progress and iron and zinc ions uptake were also regulated in 19ak-treated C. albicans cells.
