Project description:Global demand for animal protein is expected to double by 2050 at a time when resource-intensive livestock production is reaching peak capacity. Cellular agriculture offers an opportunity to meet the increasing demand for animal products, but the technology is currently limited by the genetic stability of immortalized cells, low culture yields, and high production costs. Here we demonstrate the spontaneous immortalization and long-term genetic stability of fibroblasts derived from the indigenous Israeli Baladi and the commercial Broiler Ross chicken breeds. Cells were adapted for growth as single-cell suspensions in animal-component free culture medium reaching cell densities of over 100x10^6 cells/mL in ATF perfusion presenting a production yield of over 33% w/v. We show that soy phosphatidylcholine, a major component of lecithin, activates PPAR, driving adipogenesis in immortalized chicken fibroblasts. Harvesting cultured adipocytes and blending them with high moisture extruded soy protein formed cultured chicken strips in which mouth feel and texture were supported by a blend of animal and plant proteins while aroma and flavor were driven by cultured animal fat. Visual and sensory analysis graded the product 4.5 out of 5.0, with over 85% percent of the study group said they are extremely likely to replace their food choice with this cultured meat product. The ability to create immortalized lines without genetic modification and the high yield process for cultured meat production presents an important steppingstone in the market realization of cultured meat.

Project description:Nanocrystalline cellulose (NCC) preparation in an integrated fractionation manner is expected to solve the problems of low yield and environmental impact in the traditional process. An integrated fractionation strategy for NCC production from wood was developed through catalytic biomass fractionation, the partial dissolution of cellulose-rich materials (CRMs) in aqueous tetrabutylphosphonium hydroxide, and short-term ultrasonication. The presented process could tolerate a high CRM lignin content of 21.2 wt % and provide a high NCC yield of 76.6 wt % (34.3 wt % of the original biomass). The increase in the CRM lignin content decreased the NCC yield, facilitated the crystal transition of NCC from cellulose I to cellulose II, and showed no apparent effects on the NCC morphology. A partial/selective dissolution mechanism is proposed for the presented strategy. This study provided a promising efficient fractionation-based method toward comprehensive and high-value utilization of lignocellulosic biomass through effective delignification and high-yield NCC production.

Project description:The chicken is an important agricultural animal and model for developmental biology, immunology and virology. Excess fat accumulation continues to be a serious problem for the chicken industry. However, chicken adipogenesis and obesity have not been well investigated, because no chicken preadipocyte cell lines have been generated thus far. Here, we successfully generated two immortalized chicken preadipocyte cell lines through transduction of either chicken telomerase reverse transcriptase (chTERT) alone or in combination with chicken telomerase RNA (chTR). Both of these cell lines have survived >100 population doublings in vitro, display high telomerase activity and have no sign of replicative senescence. Similar to primary chicken preadipocytes, these two cell lines display a fibroblast-like morphology, retain the capacity to differentiate into adipocytes, and do not display any signs of malignant transformation. Isoenzyme analysis and PCR-based analysis confirmed that these two cell lines are of chicken origin and are free from inter-species contamination. To our knowledge, this is the first report demonstrating the generation of immortal chicken cells by introduction of chTERT and chTR. Our established chicken preadipocyte cell lines show great promise as an in vitro model for the investigation of chicken adipogenesis, lipid metabolism, and obesity and its related diseases, and our results also provide clues for immortalizing other avian cell types.

Project description:For a subpopulation of asthmatics, symptoms persist even with high doses of glucocorticoids. Glucocorticoids reduce the levels of the proinflammatory and fibrogenic cytokine, granulocyte-macrophage colony-stimulating factor (GM-CSF) produced by human cultured airway smooth muscle (ASM). We have contrasted the effects of a synthetic glucocorticoid, dexamethasone, on thrombin- and IL-1alpha-stimulated GM-CSF production in human ASM cells. Although IL-1alpha stimulated three-fold higher levels of GM-CSF mRNA and protein compared to thrombin, dexamethasone concentration-dependently reduced IL-1alpha-stimulated GM-CSF more potently and to a greater extent than the response to thrombin. This pattern of glucocorticoid regulation was also observed at the GM-CSF mRNA level and was reproduced with other glucocorticoids such as fluticasone propionate. IL-1alpha and thrombin stimulated NF-kappa B-dependent luciferase expression equally. Dexamethasone treatment reduced luciferase expression stimulated by both IL-1alpha and thrombin. The GM-CSF mRNA half life was markedly prolonged by IL-1alpha compared to thrombin. This IL-1alpha-induced GM-CSF mRNA stability was prevented by either dexamethasone or the p38(MAPK) inhibitor, SB203580, neither of which influenced GM-CSF mRNA stability in thrombin-treated cells. Dexamethasone inhibited p38(MAPK) phosphorylation in IL-1alpha-stimulated ASM, whereas thrombin does not stimulate p38(MAPK) phosphorylation. These data suggest that the mechanism underlying the greater potency and efficacy of glucocorticoids in reducing GM-CSF synthesis stimulated by IL-1alpha depends on inhibition of the involvement of p38(MAPK)-induced increases in GM-CSF message stability.
