Global mapping of PfCDP occupany on the genome of P. falciparum

ABSTRACT: We identified wew chromodomain containing protein that binds to H3K9me3 mark in P. falciparum. To identify its global positioning on the genome of P. falciparum, we perfromed ChIP sequencing using inhouse generated anti-PfCDP antibody. The sheared chromatin was prepared from the P. falciparum culture and subjected for ChIP followed by library preparation from the ChIP DNA and were subjected to sequencing using the HiSeqIllumina platform. The PfCDP binding sites were determined after sequence alignment and normalization with input sequence. We have found selective enrichment of PfCDP protein on the virulence genes of P. falciparum. Collectively, this study reports chromodomain binds to epigenetic mark H3K9me3 and regulats the subsets of virulence genes expression in human malaria parasite P.falciparum.
