Next Generation Sequencing (NGS) of brain BRAFV600E/pAkt/Trp53KO- and control-derived tissue

ABSTRACT: Purpose: Usage of next generation sequencing (NGS) to quantify the expression levels of the overall transcriptome from brain BRAFV600E/pAkt/Trp53KO tumors and and compare it with the contralateral brain control derived from mice. Methods: Tumor tissues and adjacent normal tissues from mouse brains were microdissected, frozen in liquid nitrogen and stored at -80 °C before RNA extraction. RNA was isolated using the RNeasy Micro Kit (Qiagen) following the manufacturer´s instructions and evaluated using a NanoDrop1000 spectrophotometer and a Qubit RNA HS Assay Kit on a Qubit 2.0 Fluorometer (Thermo Fisher Scientific). Conclusion: Comparison between the transcriptome of tumor and control brain tissue rise the presence of differentially expressed genes related to immune system, inflamation, proliferation, invasion, synaptic processes, etc.
