Human lung cancer cell line A549 +/- CD43 knockdown

ABSTRACT: The human lung cancer cell line A549 with high CD43 expression was stably trasfected with a vector expressing a non-functional shRNA or 3 of these same vectors each expressing a different shRNA targeting human CD43 mRNA. The transcriptomes of these two daughter lines were then compared by differential microarray analysis. The generation and functional characteristics of these two daughter cell lines are described by Fu et al., in the International Journal of Cancer, volume 132, pages 1761-1770, published in 2013.

Project description:We discovered a spontaneous differentiation path characterized by CD43 and c-Kit in the AE9a murine model, where the leukemia-initiating potential is lost while leukemic cells display features of erythroid maturation. Therefore, we sorted out CD43++c-Kit+, CD43+c-Kit- and CD43-c-Kit- AE9a leukemic cells for RNA-sequencing analyses to determine the underlying transcriptome.

Project description:We report a novel immune molecule, CD43, that can regulate the tumor immune microenvironment (TIME) and serves as a promising target for CRC immunotherapy. The correlation of CD43 expression with CRC patient prognosis was revealed by public data analysis. CD43 KO CRC cell lines were generated by CRISPR-CAS9 technology, and a syngenetic murine CRC model was established to investigate the in vivo function of CD43.To elucidate the TIME more precisely, tumors were harvested at Day 21 postinoculation in vivo, and single-cell suspensions of tumor tissues were generated for transcriptome sequencing.

Project description:Overexpression of transcription factor Sox17 in human ES cells-derived endothelial cells and hematopoietic cells enhances expansion of hemogenic endothelium-like cells. Human ES cells were differentiated for 6 days, 8 days or 12 days in EBs, then CD34+CD43-CD45- endothelial cells, CD34+CD43+CD45- pre-hematopoietic progenitor cells (HPCs) or CD34+CD43+CD45+ HPCs were isolated by fluorescence activated cell sorting (FACS) and subjected to a microarray analysis.M-cM-^@M-^@Some samples were plated onto OP9 cells after the isolation by FACS, and transduced with the 4OH-tamoxifen-inducible 1M-CM-^WFLAG-tagged Sox17-ERT retrovirus. The cells were cultured with 4OH-tamoxifen. CD34+CD43+CD45low hemogenic endothelium-like cells expanded by Sox17-ERT were collected by magnetic-activated cell sorting (MACS) and subjected to a ChIP-chip analysis.
