Distinct Functional Properties of Perinatal vs Adult Adipose Progenitors

ABSTRACT: We previously identified functional distinct sub-popultions in adult male gWAT Pdgfrb+ cells utilizing the MuralChaser lineage tracking system and Single-cell RNA-seq. To examine the cellular altas and developmental aspects of male gWAT, we performed Single-cell RNA-seqs on different developing stage of gWAT (P3,P7 and 5-week). To examine the mesothelial origin hypothesis of gWAT, we also performed Single-cell RNA-seq on P7 gWAT associated mesothelial cells.

Later in this related submission, we performed single-cell RNA-seqs on different developing stage of gWAT (P3, P7 and 5-week). We identified 4 sub-populations of Pdgfrb+ cells. Bulk-mRNA were conducted to these sub-populations in order to understand their transcriptomic profiles.

Single cell RNA-sequencing of visceral adipose tissue Pdgfrβ+ cells

Project description:We previously derived a doxycycline-inducible (Tet-On) lineage-tracing model that allows for the indelible labeling of Pdgfrb-expressing perivascular cells in adipose tissue of adult mice (PdgfrbrtTA; TRE-Cre; Rosa26RmT/mG; herein, “MuralChaser mice”). Prior to exposing animals to doxycyline, all cells within the stromal-vascular fraction (SVF) of adult gonadal WAT (gWAT) express membrane tdTomato from the Rosa26 locus. Following 9 days of exposure to doxycycline-containing chow diet, Cre-mediated excision of the loxP-flanked tdTomato cassette occurs in Pdgfrb-expressing cells, and membrane-bound GFP (mGFP) expression is constitutively activated. We set out to test the hypothesis that Pdgfrb-expressing perivascular cells in gonadal visceral WAT of adult mice are heterogeneous, with subpopulations harboring functionally distinct phenotypes. To this end, we performed single cell RNA-sequencing of mGFP+ cells isolated from gWAT of lean (chow fed) 8 weeks-old male MuralChaser mice following 9 days of doxycycline exposure.

Project description:We report the H3K9me2 distribution profile with ChIP sequencing of postnatal male germ cells. Histone modification levels are dynamically controlled during mammalian spermatogenesis. We found that H3K9 demethylases, Jmjd1a and Jmjd1b catalyze H3K9 demethylation in prospermatogonia. Combined loss of Jmjd1 enzymes disturbed prospermatogonia to spermatogonia transition in mice. To examine a role of Jmjd1 in prospermatogonia to spermatogonia transition, we performed RNA-seq and ChIP-seq analyses using postnatal germ cells at P3 and P7.

Gene expression signature of cerebellum hypoplasia in a mouse model of Down syndrome (Part I).

Project description:We designed a large scale gene expression study in Ts1Cje mice between P0 and P10 in order to measure the effects of trisomy 21 on a large number of samples (56 in total) in a tissue that is affected in Down syndrome (the cerebellum) and to quantify the defect during development in order to correlate gene expression changes to the phenotype observed. Keywords: Down syndrome, Ts1Cje, cerebellum, development, hypoplasia We analyzed gene expression in the cerebellum of Ts1Cje and euploid mice at P0, P3, P7 and P10 using pangenomic two colors microarrays containing 25 344 probes representing approximately 15 574 mouse genes. 56 samples from individual cerebellum were hybridized on 28 microarrays. On each microarray we hybridized a Ts1Cje sample versus an euploid sample and always a male versus a female. In addition on the same microarray we always compared samples from mice of the same age or with a maximum difference of 4 days (P0 versus P3, P3 versus P7 or P7 versus P10).

Project description:To characterize the sequence of events associated with RasV12 immortalization of Drosophila embryonic cells, we generated transcriptional time series during cell line establishment, from primary cultures until passage (P) 19.
