Role of Zfp276 in oligodendrocyte differentiation

ABSTRACT: To identify target genes of the Zfp276 protein in oligodendrocytes in a genome-wide and unbiased approach, we performed RNA-Seq studies on primary mouse oligodendroglial cells (1°OGCs) that were differentiated 3 days in vitro. We compared samples generated from forebrains of mice that carried Sox10::Cre-mediated deletion of exons 2-5 of the Zfp276 gene (Zfp276cko) or Zfp276 fl/fl mice that were negative for the Cre-transgene (Ctrl). Each biological replicate was generated from 3 individuals of the same genotype at postnatal day P2-P4. Among others we detected substantial changes in the expression of genes associated with oligodendrocyte differentiation and myelination.
