Project description:Germ cells are essential to pass DNA from one generation to the next. In human reproduction, germ cell development begins with the specification of primordial germ cells (PGCs) and a failure to specify PGCs leads to human infertility. Recent studies have revealed that the transcription factor network required for PGC specification has diverged in mammals, and this has a significant impact on our understanding of human reproduction. Here, we reveal that the Hominidae-specific Transposable Elements (TEs) LTR5Hs, may serve as TEENhancers (TE Embedded eNhancers) to facilitate PGC specification. LTR5Hs TEENhancers become transcriptionally active during PGC specification both in vivo and in vitro with epigenetic reprogramming leading to increased chromatin accessibility, localized DNA demethylation, enrichment of H3K27ac, and occupation of key hPGC transcription factors. Inactivation of LTR5Hs TEENhancers with KRAB mediated CRISPRi has a significant impact on germ cell specification. In summary, our data reveals the essential role of Hominidae-specific LTR5Hs TEENhancers in human germ cell development.

Project description:The in vitro reconstitution of human germ-cell development provides a robust framework for clarifying key underlying mechanisms. Here, we explored transcription factors (TFs) that engender the germ-cell fate in their pluripotent precursors. Unexpectedly, SOX17, TFAP2C, and BLIMP1, which act under the BMP signaling and are indispensable for human primordial germ-cell-like cell (hPGCLC) specification, failed to induce hPGCLCs. In contrast, GATA3 or GATA2, immediate BMP effectors, combined with SOX17 and TFAP2C, generated hPGCLCs. GATA3/GATA2 knockouts dose-dependently impaired BMP-induced hPGCLC specification, whereas GATA3/GATA2 expression remained unaffected in SOX17, TFAP2C, or BLIMP1 knockouts. In cynomolgus monkeys, a key model for human development, GATA3, SOX17, and TFAP2C were co-expressed exclusively in early PGCs. Crucially, the TF-induced hPGCLCs acquired a hallmark of bona fide hPGCs to undergo epigenetic reprogramming and mature into oogonia/gonocytes in xenogeneic reconstituted ovaries. By uncovering a TF circuitry driving the germ line program, our study provides a paradigm for TF-based human gametogenesis.

A Signaling and Transcription Architecture for the Specification of Human Germ Cell Lineage

Project description:The mechanism for human germ-cell specification, which sets out a complex program generating spermatozoa or oocytes, remains largely unknown. We established a system for inducing human primordial germ cell-like cells (hPGCLCs) from induced pluripotent stem cells (hiPSCs) via incipient mesoderm-like cells (iMeLCs). Here, we show that EOMESODERMIN (EOMES) elevates in iMeLCs through WNT signaling and is essential for activating SOX17, a key driver for hPGC(LC) specification, with the duration/dosage of the WNT signaling/EOMES expression dictating the germ-cell competence. Upon hPGCLC induction, BMP signaling activates TFAP2C independently from SOX17, and SOX17 and TFAP2C instate the hPGCLC program, including BLIMP1 expression, in an interdependent fashion. The hPGC(LC) specification program is highly consistent with the monkey program, but is divergent from the mouse one regarding key transcription factors and their hierarchy of actions. These findings delineate evolutionary divergence of mammalian germ-cell specification, providing a foundation for further human germ-cell development in vitro.
