Project description:Dilated cardiomyopathy (DCM), a myocardial disorder that can result in progressive heart failure and arrhythmias, is defined by ventricular chamber enlargement and dilatation, and systolic dysfunction. To decipher the basis for the cardiac pathology in titin-mutated patients, we investigated the hypothesis that induced Pluripotent Stem Cell (iPSC)- derived cardiomyocytes (iPSC-CM) generated from patients, recapitulate the disease phenotype.Our findings show that the mutated cardiomyocytes from DCM patients recapitulate abnormalities of the inherited cardiomyopathies.

ATAC-seq in human iPSC-derived cardiomyocytes (CMs)

Project description:The human iPSC line H19101 was differentiated in vitro into cardiomyocytes using a 20-day differentiation protocol (Burridge et al. 2014 PMID 24930130 and Montefiori et al 2018 PMID 29988018 ). 50,000 cardiomyocytes were used in each ATAC-seq experiment. 8 replicates were pooled to obtain the final peak file.

Age-Dependent Maturation of iPSC-CMs Leads to the Enhanced Compartmentation of ?2AR-cAMP Signalling.

Project description:The ability to differentiate induced-pluripotent stem cells into cardiomyocytes (iPSC-CMs) has opened up novel avenues for potential cardiac therapies. However, iPSC-CMs exhibit a range of somewhat immature functional properties. This study explored the development of the beta-adrenergic receptor (?AR) pathway, which is crucial in regulating contraction and signifying the health and maturity of myocytes. We explored the compartmentation of ?2AR-signalling and phosphodiesterases (PDEs) in caveolae, as functional nanodomains supporting the mature phenotype. Förster Resonance Energy Transfer (FRET) microscopy was used to study the cyclic adenosine monophosphate (cAMP) levels in iPSC-CMs at day 30, 60, and 90 following ?AR subtype-specific stimulation. Subsequently, the PDE2, PDE3, and PDE4 activity was investigated using specific inhibitors. Cells were treated with methyl-?-cyclodextrin (M?CD) to remove cholesterol as a method of decompartmentalising ?2AR. As iPSC-CMs mature with a prolonged culture time, the caveolae density is increased, leading to a reduction in the overall cytoplasmic cAMP signal stimulated through ?2AR (but not ?1AR). Pan-phosphodiesterase inhibition or caveolae depletion leads to an increase in the ?2AR-stimulated cytoplasmic cAMP. Moreover, with time in culture, the increase in the ?AR-dependent cytoplasmic cAMP becomes more sensitive to cholesterol removal. The regulation of the ?2AR response by PDE2 and 4 is similarly increased with the time in culture. We conclude that both the ?2AR and PDEs are restricted to the caveolae nanodomains, and thereby exhibit a tighter spatial restriction over the cAMP signal in late-stage compared to early iPSC-CMs.

Genome-wide estrogen-related receptor gamma (ERRg) occupancy in human iPS cell-derived cardiomyocytes (hiPSC-CMs)

Project description:Estrogen-related receptor gamma (ERRg) has been shown to control gene expression involved in a broad range of mitochondrial energy metabolism including oxidative phosphorylation, TCA cycle, and fatty acid oxidation. However, ERRg direct targets were not identified in cardiomyocytes. With ERRg ChIP-seq, we found ERRg peaks on the promoter regions of mitochondrial energy metabolic genes as expected. Besides, ERRg extensively distributed the promoter regions of cardiac contractile, ion channels and Ca2+ handling protein genes. Surprisingly, the peaks also were found on non-cardiomyocyte genes and genes expressed in early-stage cardiomyocytes.
