Extracellular ectonucleotidases are differentially regulated in murine tissues and human polymorphonuclear leukocytes during sepsis and inflammation

ABSTRACT: In the present studies we identified NTPDase8 a top downregulated gene in the liver of mice submitted to the cecal ligation-induced sepsis. Immunohistochemical analysis confirmed the decrease on NTPDase8 expression at the protein level. In vitro mechanistic studies using HepG2 hepatoma cells demonstrated that IL-6 but not TNF, IL-1b, bacteria or lipopolysaccharide are able to suppress NTPDase8 gene expression.

Project description:Interleukin-1B (IL-1B) is pathologically activated by inflammasome-associated caspase-1 in rare autoinflammatory conditions and in wide-spread diseases. Therefore, IL-1B activity must be fine-tuned to enable anti-microbial responses whilst limiting collateral damage. Here we report that, relative to other inflammasome components, IL-1B is rapidly turned over by the proteasome and this correlates with its decoration by K11-, K63- and K48-linked ubiquitin chains. This IL-1 degradation signal is not only a feature of inflammasome priming but is also triggered upon inflammasome activation. We demonstrate that IL-1 K133 is modified by ubiquitin and forms a salt bridge with IL-1B D129. Loss of IL-1 K133 ubiquitylation, or disruption of the K133:D129 electrostatic interaction, stabilizes IL-1Bprotein levels. Accordingly, IL-1B K133R/K133R mice display increased precursor IL-1Bupon inflammasome priming and increased bioactive IL-1B following inflammasome activation, both in vitro and following LPS injection in vivo. These findings reveal new mechanisms for limiting IL-1B activity and safeguarding against damaging inflammation.

Long non-coding RNAs are central regulators of the IL-1b-induced inflammatory response in human lung fibroblasts

Project description:There is accumulating evidence to indicate that long non-coding RNAs (lncRNAs) are important regulators of the inflammatory response. In this report, we have employed next generation sequencing to identify 14 lncRNAs that are differentially expressed in human lung fibroblasts following the induction of inflammation using interleukin-1b (IL-1b). Knockdown of the two most highly expressed lncRNAs, IL7AS and MIR3142HG, showed that IL7AS negatively regulated IL-6 release whilst MIR3142HG was required for IL-8 and CCL2 release. Parallel studies in fibroblasts derived from patients with idiopathic pulmonary fibrosis showed similar increases in IL7AS levels, that also negatively regulate IL-6 release. In contrast, IL-1b-induced MIR3142HG expression was reduced by 8-fold in IPF fibroblasts, with the consequence being that MIR3142 knockdown showed no effect upon IL-8 and CCL2 release. In summary, we have catalogued those lncRNAs that are differential expression following IL-1b-activation of human lung fibroblasts and shown that these regulate the inflammatory response.

Project description:Inflammation resolution is critical for sepsis induced acute lung injury (ALI) recovery. Interleukin-36 receptor (IL-36R) is a potent anti-inflammatory factor. However, its role in ALI resolution remains unclear. We investigated the effects of IL-36R during the ALI resolution process in a murine cecal ligation and puncture (CLP)-induced ALI model. Knockout IL-36R signaling aggravates CLP-induced lung injury, as manifested by elevated bacterial load and increased neutrophils recruitment to the lung. Thereafter, we used IL-36R knockout mice to discern the source cell of IL-36R during ALI. We found that IL-36R is predominantly generated by epithelial cells during the ALI process. Furthermore, we sorted lung epithelial cells on the ALI process. IL-36R-specific loss in epithelial cells leads to apoptosis through NF-κB pathway. Together, our findings identify molecules that are likely involved in sepsis induced lung injury that may inform biomarker and therapeutic development.
