PanCancer Immune Nanostring analysis of PBMCs stimulated with Innate-like T-cell agonists

ABSTRACT: We aimed to study the function of NKT, MAIT and V[gamma]9V[delta]2 T-cells in a population of people with cancer. To select appropraite immune parameters to study, we wanted to compare and contrast the expression of immune-related genes in human PBMCs following NKT, MAIT and V[gamma]9V[delta]2 T-cell activation. Aim: to compare the expression of immune-related genes in human PBMCs following NKT-cell activation with alpha-galactosylceramide, MAIT-cell activation with 5-amino-6-d-ribitylaminouracil/Methyl glyoxal, and V[gamma]9V[delta]2 T-cell activation with Bromohydrin pyrophosphate (BrHPP).

Microarray expression profiling of NKT and MAIT subsets in different organs of B6-MAIT/CAST mice.

Project description:MAIT cells (MAITs) represent an abundant T lymphocyte subset with unique specificity for microbial metabolites presented by the MHC-1b molecule, MR1. MAIT conservation along evolution indicates important, non-redundant functions, but their low frequency in mice has hampered their detailed characterization. Here, we performed a transcriptomic analysis of murine MAITs in comparison with NKT subsets and with mainstream T cells in spleen and peripheral organs of B6-MAIT/CAST mice expressing a Rorc-GFP transgene. MAIT and NKT cells have been FACS-sorted after tetramer staining (MR1:5-OP-RU Tet+ for MAIT, CD1d:PBS57Tet+ for NKT), and 1/17 subsetting based on the expression of Rorc.

Activation of natural killer T cells promotes M2 macrophage polarization in adipose tissue and improves systemic glucose tolerance via the IL-4/STAT6 signaling axis in obesity

Project description:Natural killer T (NKT) cells are important therapeutic targets in various disease models and under clinical trials for cancer patients. However, their function in obesity and type 2 diabetes remains unclear. Our data show that adipose tissues of both mice and humans contain a population of type-1 NKT cells, whose abundance decreases with increased adiposity and insulin resistance. Although loss-of-function of NKT cells had no effect on glucose tolerance in animals with prolonged high-fat diet (HFD) feeding, activation of NKT cells by lipid agonist α-galactosylceramide (αGalCer) enhances alternative macrophage polarization in adipose tissue and improves glucose homeostasis in animals at different stages of obesity. Furthermore, the effect of NKT cells is largely mediated by the IL-4/STAT6 signaling axis in obese adipose tissue. Thus, our data identifies a novel therapeutic target for the treatment of obesity-associated inflammation and type-2 diabetes. Wild type and CD1d1 antigen (CD1d) knockout mice were fed a high fat diet for 4 days. On day 0 and 2 mice were injected intraperitoneally with α-galactosylceramide (αGalCer) or vehicle. On day 4 mice were killed and gene expression was profiled in epididymal adipose tissue.

Natural killer T-cell characterization through gene expression profiling

Project description:Natural killer T (NKT) cells are a distinct lymphocyte lineage thought to operate primarily at the interface between the innate and adaptive immune response. Yet, their unique role in the immune system remains elusive. Whilst NKT cells show high similarities to other cells of the innate and adaptive immune system, they express unique functional features such as rapid, concomitant production of Th1 and Th2 cytokines upon TCR ligation. In order to portray gene expression of NKT cells and to analyze their complete functional potential, we performed comparative microarray analyses of naive NKT cells, naive NK cells as well as naive conventional CD4+ T cells (Th, CD4+CD25–) and naive regulatory CD4+CD25+ T cells (Treg ). Furthermore, we compared the gene expression profiles of naive versus alpha-galactosylceramide activated NKT cells to elucidate the gene set rapidly expressed upon activation. We describe profound gene expression differences between the different cell types as well as between naive and activated NKT cells allowing the identification of a unique gene expression profile of NKT cells. In addition to known NKT cell specific markers, a high number of genes were expressed and detected which had not been attributed to NKT cells. Notably, our analyses reveals that NKT cells are not only of Th1 and Th2 type but also fulfil criteria of Th17 cells. Hence, our data provide new insight into the genetic décor of NKT cells which will facilitate a better understanding of their versatile role during the immune response. The effect of NKT cell activation on the transcriptome of intestinal epithelial cells

Project description:Intestinal homeostasis is maintained through the combined functions of epithelial and immune cells that collaborate to preserve the integrity of the intestinal barrier. However, the mechanisms by which immune cell populations regulate intestinal epithelial cell (IEC) homeostasis remain unclear. Here, we use a multi-omics approach to study the immune-epithelial crosstalk and identify CD1d-restricted Natural Killer T (NKT) cells as regulators of IEC biology. We find that NKT cells are abundant in the proximal small intestine and show hallmarks of activation at steady state. Subsequently, NKT cells regulate the survival and the transcriptional and cellular composition landscapes of IECs in intestinal organoids, through mechanisms dependent on IFN-γ and IL-4 secretion by NKT cells but independent of the expression of CD1d on IECs. In vivo, lack of NKT cells results in an increase in IEC turnover, while NKT cell activation leads to IFN-γ-dependent epithelial apoptosis. Our findings propose NKT cells as potent producers of cytokines that contribute to the regulation of IEC homeostasis.
