Project description:IL-2 mutant (F42K) is a variant of IL-2 that binds preferentially to the lower affinity IL-2RÎ²Î³ and has minimal binding to CD25, on Tregs, effector NK and T-cell subsets. Our recent study has uncovered that in contrast to WT IL-2, F42K did not efficiently induce the expansion of highly suppressive ICOS+ Tregs in PBMC from healthy controls and melanoma patients whereas it did promote the expansion of NK cells. We used microarray to study the differences of gene profiles induced by wild-type (WT) IL-2, F42K and IL-15 when treated on freshly isolated human peripheral blood mononuclear cells (PBMC) from healthy control. Human peripheral blood mononuclear cells (PBMC) were isolated from buffy coat and were stimulated with 0.4nM WT IL-2, F42K or IL-15 for 24 hr. Total RNA was extracted from each WT IL-2, F42K and IL-15 treated PBMCs and Sense RNA was reverse-transcribed via RT reaction into DNA and labeled with biotin using Affymetrix labeling kits and hybridized on human Affymetrix microarray chip.

CD158a+/CD158b+ NK Cell Imbalance Correlates with Hypertension in Patients with Pre-eclampsia

Project description:Background: Preeclampsia, a pregnancy complication with hypertension and proteinuria, seriously threats the health and lives of the mother and the baby. The role of peripheral natural killer cells (NK cells) in the pre-eclampsia is unclear. Methods: Flow cytometry was performed to detect the expression of CD158a (KIR2DL1) and CD158b (KIR2DL2/3) in peripheral NK cells of non-pregnant healthy women (HW), healthy pregnant women (HP), and patients with pre-eclampsia (PE). Differentially expressed genes (DEGs) in CD158a+ and CD158b+ NK cells were identified by RNA-sequencing and real-time PCR. Protein array analysis was used to identify altered protein levels in the serum of study participants. Findings: CD158a+ NK cell numbers were significantly increased in the peripheral blood of patients with pre-eclampsia while the number of CD158b+ NK cells was significantly reduced. The percentage of CD158a+ NK cells was positively correlated with systolic blood pressure while the percentage of CD158b+ NK cells was negatively correlated with systolic blood pressure. RNA-seq and real-time PCR showed that the expression of ERAP2 and GCH1, the genes that regulate blood pressure and angiogenesis, was decreased in CD158a+ compared to CD158b+ NK cells. Interpretation: CD158a+ and CD158b+ NK cell numbers are altered in the peripheral blood of patients with pre-eclampsia compared to healthy individuals, which is associated with the increase in blood pressure.

Gene expression profiling of human NK cells

Project description:In order to identify genes expression in different types of human NK cells, we sorted normal decidual NK cells during the first trimester, NK cells from peripheral blood and cord blood for analysis using the Agilent Whole Human Genome Microarray. Samples were collected from healthy adult donors after obtaining informed conset according to the Ethics Committee of the University of Science & Technology of China. CD56+CD3- NK cells have been sorted from normal decidua, cord blood and peripheral blood by FACS Aria. Then Whole Human Genome Microarray was employed as a discovery platform to identify genes differentially expressed in each types of NK cells.

Diversity of peripheral blood human NK cells identified by single cell RNA sequencing

Project description:Human Natural Killer (NK) cells in peripheral blood perform many functions but classification of specific subsets has been a long-standing problem. Here, we report single-cell RNA sequencing of NK cells from healthy CMV-negative donors, comparing gene expression in unstimulated and IL-2 activated cells. Unsupervised clustering identified seven NK cell subsets. Three resembled well-described populations, CD56brightCD16-, CD56dimCD16+CD57- and CD56dimCD16+CD57+. CD56dimCD16+CD57- cells sub-divided to include a population with higher chemokine mRNA and increased frequency of KIR expression. Three novel human blood NK cell populations were identified as: a population of type I interferon responding NK cells which were CD56neg; a population exhibiting an in-vivo cytokine-induced memory-like phenotype, including increased granzyme B mRNA in response to IL-2, and finally, a small population, with low ribosomal expression, down-regulation of oxidative phosphorylation and high levels of immediate early response genes indicative of cellular activation. Human cytomegalovirus positive donors also included a higher frequency of adaptive NK cells. Together, these data establish an unexpected diversity in blood NK cells and provide a new framework for analysing NK cell responses in health and disease.
