Project description:To investigate the molecular mechanisms associated with initial dose-related events that are linked to the development of liver tumours: liver growth; cell proliferation; changes in histopathology such as hypertrophy Experiment Overall Design: We selected dose levels of phenobarbital to give a carcinogenic (1000ppm) and two non-carcinogenic doses (50 and 500ppm). This also gave two doses that increased liver weights (500 and 1000ppm) and one that did not (50ppm). Alterations to liver growth and proliferation are complete after 14 days of dietary treatment and so we chose intermediate time points of 1,3 7 and 14 days of treatement. Small sections of the left lateral lobe of the liver were processed to give three biological replicates selected from the 5 treated animals in each group, total RNA purified and mRNA levels measured using Affymetrix Rat Genome 230 v2 microarrays. In addition we also measured a braod range of standard histopathological, clincal chemsitry and metabolomic endpoints.

Project description:To provide insights into the mode of action for Ni3S2 lung carcinogenicity by examining gene expression changes in target cells after inhalation exposure. Gene expression changes were determined in micro-dissected lung broncho-alveolar cells from Fischer F344 rats following inhalation of Ni3S2 at 0.0, 0.04, 0.08, 0.15, and 0.60 mg/m3 (0.03, 0.06, 0.11, and 0.44 mg Ni/m3) for one and four weeks (6 hours per day, 5 days per week). Results: Broncho-alveolar lavage fluid evaluation and lung histopathology provided evidence of inflammation only at the two highest concentrations, which were similar to those tested in the 2-year bioassay. The number of statistically significant up- and down-regulated genes decreased markedly from one to four weeks of exposure, suggesting adaptation. Cell signal pathway enrichment at both time-points primarily reflected responses to toxicity, including inflammatory and proliferative signaling. While proliferative signaling was up-regulated at both time points, some inflammatory signaling reversed from down-regulation at 1 week to up-regulation at 4 weeks. Conclusions: These results support a mode of action for Ni3S2 carcinogenicity driven by chronic toxicity, inflammation and proliferation, leading to mis-replication, rather than by direct genotoxicity. Benchmark dose (BMD) analysis identified the lowest pathway transcriptional BMD exposure concentration as 0.026 mg Ni/m3, for apoptosis/survival signaling. When conducted on the basis of lung Ni concentration the lowest pathway BMD was 0.64 µg Ni/g lung, for immune/inflammatory signaling. Implications: These highly conservative BMDs could be used to derive a point of departure in a nonlinear risk assessment for Ni3S2 toxicity and carcinogenicity.

We analyzed MR-induced changes in gene expression using pooled RNA from liver of rats fed either a control purified amino acid diet (DL-methionine content of 0.86%) (CON) or a methionine-restricted diet (DL-methionine content of 0.172%)(MR). Rats were fed Purina rodent diet 5001 until 32 days of age and were then randomly assigned to be fed CON diet or MR diet for 20 months.

Project description:Aging is associated with a substantial decline in social behavior, whereas positive social interaction can improve overall health in aged individuals. In laboratory rodents, manipulations of the social environment across the lifespan have been shown to affect social behavior. Therefore, we examined the effects of long-term (5-6?weeks) housing conditions (alone, with one adult, or with two adults) on social behavior and the expression of neuroinflammation-related genes as well as oxytocin receptor (OXTR) gene expression in brain areas associated with social behavior regulation in aged male and female Fischer (F) 344 rats. Single-housed males and females exhibited increased social investigation, relative to pair-housed rats (one aged and one adult). Triple-housed (one aged and two adults) aged males exhibited lower levels of social investigation, relative to triple-housed aged females. Aged females were more socially active that their male counterparts. Although social housing condition significantly affected social behavior in males, it had no impact on cytokine gene expression in the paraventricular nucleus of hypothalamus (PVN), bed nucleus of the stria terminalis (BNST) or medial amygdala (MeA). However, in triple-housed aged females, who exhibited social behavior comparable to their single- and pair-housed counterparts, there was a significant increase in the expression of IL-1? and IL-6 mRNA in the MeA. No changes in cytokine gene expression were observed in the PVN or BNST, indicating that the increased expression of cytokines in the MeA was not a result of a generalized increase in neuroinflammation. Single-housed males and females exhibited elevated OXTR gene expression in the BNST. Taken together, these data indicate that manipulations of the social environment in late aging significantly influenced social interactions with a novel partner and gene expression in social behavior circuits and that these effects are sex-specific.

Project description:Recent evidence suggests that older adults may be a sensitive population with regard to environmental exposure to toxic compounds. One source of this sensitivity could be an enhanced variability in response. Studies on phenotypic differences have suggested that variation in response does increase with age. However, few reports address the question of variation in gene expression as an underlying cause for increased variability of phenotypic response in the aged. In this study, we utilized global analysis to compare variation in constitutive gene expression in the retinae of young (4 months), middle-aged (11 months), and aged (23 months) Fischer 344 rats. Three hundred and forty transcripts were identified in which variance in expression increased from 4 to 23 months of age, while only 12 transcripts were found for which it decreased. Functional roles for identified genes were clustered in basic biological categories including cell communication, function, metabolism, and response to stimuli. Our data suggest that population stochastically induced variability should be considered in assessing sensitivity due to old age.
