Highly functional intestinal monolayer through organoids derived from human iPS cells for drug discovery research

Project description:Human induced pluripotent stem (iPS) cell-derived enterocyte-like cells (ELCs) are expected to evaluate intestinal absorption and metabolism of orally administered drugs. However, it was difficult to generate large amounts of ELCs with high quality because they cannot proliferate and be passaged. In this study, we established the organoids from ELCs (ELC-org), which could be passaged and maintained for more than a year. When ELC-org were dissociated into single cells and seeded on cell culture inserts (ELC-org-mono), they formed a tight monolayer in 3 days. Both ELC-org and ELC-org-mono were composed exclusively of epithelial cells, being nearly 100% of EpCAM-positive. Gene expressions of many drug-metabolizing enzymes and drug transporters were enhanced in ELC-org-mono compared to those in ELCs and ELC-org. Their levels were similar to those in human adult intestine. The CYP3A4 activity level in ELC-org-mono was comparable or higher than that in primary cryopreserved human small intestinal cells and their expression level was up-regulated by inducers such as rifampicin. ELC-org-mono had the efflux activities of P-gp and BCRP. Importantly, ELC-org-mono cultured for more than a year maintained high intestinal functions. Even when the cryopreserved organoid-derived cells were directly seeded and cultured on cell culture inserts, they maintained high intestinal functions similar to the cells without cryopreservation. RNA-seq analysis showed that ELC-org-mono were more mature as intestinal epithelial cells than ELC and ELC-org. Therefore, we have successfully improved the function and convenience of ELCs by utilizing organoid technology.

Generation of human cerebral organoids with a structured outer subventricular zone

Project description:Outer radial glia (oRG) emerged during mammalian evolution as cortical progenitor cells that directly support the development of an enlarged outer subventricular zone (oSVZ) and, in turn, the expansion of the neocortex. The in vitro generation of oRG is essential to model and investigate the underlying mechanisms of human neocortex development and expansion. By activating the STAT3 pathway using LIF, which is not produced in guided cortical organoids, we developed a cerebral organoid differentiation method from human pluripotent stem cells (hPSCs) that recapitulates the expansion of a progenitor pool into the oSVZ. The structured oSVZ is composed of progenitor cells expressing specific oRG markers such as GFAP, LIFR, HOPX and closely matching human oRG in vivo. In this microenvironment, cortical neurons showed faster maturation with enhanced metabolic and functional activity. Incorporation of hPSC-derived brain vascular LIF-producing pericytes in cerebral organoids mimicked the effects of LIF treatment. These data indicate that the cellular complexity of the cortical microenvironment, including cell types of the brain vasculature, favors the appearance of oRG and provides a platform to routinely study oRG in hPSC-derived brain organoids.

Expression data from C3(1)Tag mammary tumors, tumor derived cell line (M6) and normal mammary tissue (FVB)

Project description:M6 cells expression a similar genetic signature as the parent tumor, C3(1)Tag tumor. The mammary tumors, and tumor cell line, are distinct from normal mammary epithelial tissue (FVB) BRCA/p53, cmyc, h2n, p53ERneg, p53ERpos, pymt and ras samples were not reported in this paper. We used microarrays to determine whether the M6 cell line retains the SV40Tag signature (91 gene signature) that is expressed in C3(1)Tag tumors but not FVB normal mammary tissue.

Chemotherapy-mediated changes in the immune cell landscape of murine triple negative breast cancer.

Project description:We performed CITE-seq (10x Genomics-based) to profile and compare the transcriptomes and cell surface expression of immune epitopes in the primary breast tumors of transgenic mice (C3-1-TAg) with or without MCT chemotherapy. We sequenced a total of six different tumor samples. We created an antibody pool consisting of 35 different antibodies and stained each sample individually with this antibody pool. Then, we stained each sample with it's own unique hashing antibody so that we could subsequently pool the samples for loading onto the 10x Chromium and later prepare one library consisting of all sight samples and finally separate each sample in silico by it's unique hashing antibody. The samples are as follows: (1) HTO1: tumor of PBS treated C3-1-TAg mouse; (2) HTO2: tumor of PBS treated C3-1-TAg mouse; (3) HTO3: tumor of PBS treated C3-1-TAg mouse; (4) HTO4: tumor of MCT treated C3-1-TAg mouse; (5) HTO5: tumor of MCT treated C3-1-TAg mouse; (6) HTO6: tumor of MCT treated C3-1-TAg mouse. The following sample comparisons were made: HTO1, HTO2, and HTO3 versus HTO4, HTO5, and HTO6.
