Pegvisomant-treated mouse prostate epithelium renal grafts

ABSTRACT: PTEN-CaP2 mouse prostate cancer cells were grafted under the kidney capsule of immunocompromised mice. After three weeks, mice were treated with 100 mg/kg pegvisomant (provided by Pfizer) or PBS vehicle 3X weekly for three weeks. Grafts were harvested three days after final treatment, flash frozen, and total RNA was extracted for sequencing.

Real-time quantitative PCR analysis of human total blood tissue from athletes or sedentary controls

Project description:Thirteen elite handball athletes and 13 sedentary controls. Three timepoints were established: T0 (baseline conditions); T8 (after 8 weeks of supplementation); and T16 (after 8 weeks in the absence of supplementation). The dietary intervention consisted of the oral administration of one daily multivitamin/mineral complex capsule (Multicentrum® Pfizer, Barcelona, Spain) before exercise during the controlled dietary intervention period. Multivitamin/mineral complex intervention adherence/compliance was defined as the percentage of all of the supplement capsules ingested throughout the study period. The expressions of a total 112 of genes were evaluated by RT-qPCR analysis with the QuantStudioTM 12K Flex Real-Time PCR System. 78 genes were finally analized.

Transcriptome data from human endocrine cells recovered from mouse grafts with pre-engraftment controls

Project description:We examined the transcriptomes of alpha and beta cells recovered from mouse sub-renal capsule grafts of human islets as compared to alpha and beta cells from the same islet preps pre-engraftment. We found that after three to four weeks in the graft the human islets were still glucose responsive, secreting insulin following glucose challenge. In addition, gene expression was largely unchanged from the pre-engraftment control samples.

mRNA aging shapes the Cap2 methylome in mammalian mRNA

Project description:The mRNA cap structure is a major site of dynamic mRNA methylation. mRNA caps exist in either the Cap1 or Cap2 form, depending on the presence of 2’-O-methylation on the first, or both the first and second transcribed nucleotide, respectively. However, the identity of Cap2-containing mRNAs and the function of Cap2 are unclear. Here we describe CLAM-Cap-Seq, a method for transcriptome-wide mapping and quantification of Cap2. We find that unlike other epitranscriptomic modifications, Cap2 can occur on all mRNAs. Cap2 is formed through a slow continuous conversion of mRNAs from Cap1 to Cap2 as mRNAs age in the cytosol. As a result, Cap2 is enriched on long-lived mRNAs. We find that large increases in the abundance of Cap1 leads to activation of RIG-I, especially in conditions where RIG-I expression is increased. The methylation of Cap1 to Cap2 markedly reduces the ability of RNAs to bind and activate RIG-I. We find that the slow Cap2 methylation rate allows Cap2 to accumulate on host mRNAs yet ensures that low Cap2 levels occur on newly expressed viral RNAs. Overall, these results reveal an immunostimulatory role for Cap1, and that Cap2 functions to reduce activation of the innate immune response.

Project description:Transcriptional changes occurring at the graft interface of auto- (Cabernet Sauvignon/ Cabernet Sauvignon) and hetero-grafted (Cabernet Sauvignon/ Riparia Gloire de Montpellier and Cabernet Sauvignon /1103 Paulsen) grapevine. Gene expression profiling was done using the Nimblegen whole genome array with 3 biological replicates of 15 pooled graft interfaces harvested 3, 7, 14 and 28 d after grafting.

Project description:Lisbon lemon trees were grafted with budwood infected with the citrus greening bacterium, 'Candidatus Liberibacter asiaticus', or control budwood, and leaf samples were collected every two weeks post graft for LC/MS analysis. This project has shotgun proteomics data for leaf samples from 5 control and 5 CLas grafted trees at the 14 week post graft timepoint.

Salicylic acid treatment and its effect on seed yield and seed molecular composition of Pisum sativum under abiotic stress

Project description:Pisum sativum (cv. Kudrnac) seeds were obtained from SEMO a.s., Smržice, Czechia. All plants used for the study grew in the experimental field plot at Veletiny, Czechia (49°02'N 17°33'E; 195 m a.s.l.). Sowing was done after the rainfall on June 1, 2022, and plants were grown in a natural environment without any additional treatment. After 35 days, the field was divided into three blocks (mock, SAt1, SAt2), each containing approxi-mately 100 plants. For five weeks, plants were weekly sprayed with 1.5 L water solution supplemented with 0.025% (v/v) Silwet L-77 (mock; K), or 100 µM salicylic acid and Silwet (final concentration as for the mock; SA-a). The third set was treated as the mock for two weeks, followed by four weeks with salicylic acid treatment (SA-b).

Project description:The fetal ovarian grafts under the kidney capsule of adult male mice undergo a partial sex-reversal showing ectopic SOX9-positive Sertoli cell-like cells around 15-20 days post-transplantation. However, the molecular bases of such masculinization of fetal ovaries in the paternal environment were unclear. We examined the temporal changes of the gene expression profiles of the grafted fetal ovaries during the partial masculinizing process in the male nude mice.
