A Robust Auxin Response Network Controls Embryo and Suspensor Development through a bHLH Transcriptional Module

Project description:Land plants can reproduce sexually by developing an embryo from a fertilized egg cell. However, embryos can also be formed from other cell types in many plant species. A key question is thus how embryo identity in plants is controlled, and how this process is modified during non-zygotic embryogenesis. The Arabidopsis zygote divides to produce an embryonic lineage and an extra-embryonic suspensor. Yet, normally quiescent suspensor cells can develop a second embryo when the initial embryo is damaged, or when response to the signaling molecule auxin is locally blocked. Here we have used auxin-dependent suspensor embryogenesis as a model to determine transcriptome changes during embryonic reprogramming. We find that reprogramming is complex and accompanied by large transcriptomic changes prior to anatomic changes. This analysis revealed a strong enrichment for genes encoding components of auxin homeostasis and response among misregulated genes. Strikingly, deregulation among multiple auxin-related gene families converged upon re-establishment of cellular auxin levels or response. This suggests a remarkable degree of feedback regulation to create resilience in auxin response during embryo development. Starting from the transcriptome of auxin-deregulated embryos, we identify an auxin-dependent bHLH transcription factor network that mediates the activity of this hormone in suppressing embryo development from the suspensor.

ABP1-TMK auxin perception mediates rapid phosphorylation for regeneration and auxin canalization

Project description:The phytohormone auxin triggers transcriptional reprograming utilizing a well-characterized nuclear perception machinery. In contrast, mechanisms underlying other auxin effects, such as auxin feed-back on its transport, rapid regulation of ion fluxes or ultrafast global phospho-response, remain enigmatic. Auxin Binding Protein 1 (ABP1) has been contested as an auxin receptor candidate since decades. Here we show that a fraction of Arabidopsis thaliana ABP1 is secreted and binds auxin specifically at the acidic pH typical for the apoplast. ABP1 and its plasma membrane-localized partner, Transmembrane Kinase 1 (TMK1) are required for auxin-induced phosphorylation of about thousand proteins. Loss-of-function abp1 alleles but not complemented lines show defects in in vitro shoot regeneration and formation of auxin-transporting channels for vasculature formation and regeneration. These results support a role of ABP1-TMK1 in cell surface auxin perception mediating global auxin phospho-response and regenerative development.

AUXIN RESPONSE FACTOR3 Integrates the Functions of AGAMOUS and Auxin to Repress Cytokinin Biosynthesis and Signaling in Floral Meristem Determinacy

Project description:Successful floral meristem (FM) determinacy is critical for the subsequent reproductive development and life cycle. Although phytohormones cytokinin and auxin interact to coordinately regulate many aspects of plant development, whether, if so how, cytokinin and auxin function in FM determinacy remain unclear. Here we show that the homeostasis of cytokinin is critical for the FM determinacy. Auxin promotes the expression of AUXIN RESPONSE FACTOR3 (ARF3) to repress cytokinin activity in FM determinacy. We found that ARF3 represses the expression of the ISOPENTENYLTRANSFERASE (IPT) family genes directly and the LONELY GUY (LOG) family genes indirectly, whose products are enzymes required for cytokinin biosynthesis. Furthermore, ARF3 directly inhibits the expression of ARABIDOPSIS HISTIDINE KINASE4 (AHK4), one of cytokinin receptor genes, to depress cytokinin activity. Genome-Wide Transcriptional Profiling reveals multiple functions of ARF3 in flower development. Consequently, ARF3 controls cell division by regulating cell cycle genes expression through cytokinin. Eventually, we show that AGAMOUS (AG) dynamically regulates the expression of ARF3 and IPTs resulting in coordinated regulation of FM maintenance and termination through cell division. Therefore, our findings reveal the molecular linkage between phytohormones auxin/cytokinin and AG in FM determinacy and shed light on the mechanisms of FM maintenance and termination.
