RNA sequencing of wild type or calcineurin-deficient mouse bone marrow-derived dendritic cells stimulated with or without lipoarabinomannan (LAM) from Mycobacterium tuberculosis Aoyama-B

ABSTRACT: To elucidate the function of calcineurin in dendritic cells (DCs) downstream of C-type lectin receptor, we established mouse model that has DC-specific calcineurin deficiency. We stimulated the bone marrow-derived DCs from these mice with lipoarabinomannan (LAM), a ligand of Dectin-2, and performed RNA sequencing. From multiple genes that showed altered expression pattern in calcineurin-deficient DCs, we found that Il10 transcription increased in the absence of calcineurin. Our further study elucidated the mechanism by which calcineurin regulates Il10 transcription in LAM activated DCs.
