Chicory: cutting the effects and the effectors for a functional food

ABSTRACT: Whole chicory flour (Chic) and three compounds of chicory roots: fructose (Fru), chlorogenic acids (CGA) and sesquiterpene lactones (STL) were separately analyzed for their health effects in mice. A whole transcriptomic analysis was conducted using Agilent DNA microarrays to investigate their nutrigenomic effects.

Project description:In Drosophila,male courtship behaviour serves as an excellent paradigm to study how innate behaviors are controlled by the nervous system. These behaviors are in large part regulated by the gene fruitless (fru). fru encodes a set of putative transcription factors that promote male sexual behaior by controlling the development of sexually-dimorphic neuronal circuitry. Little is known about how Fru proteins function at the level of transcriptional regulation. To characterize the roles of fru sex-specific isoforms in specifying male behavior, we generated novel isoform-specific mutants, and used a genomic approach to identify direct Fru isoform targets during development. We demonstrate that all Fru isoforms directly target genes involved in the development of the nervous system, with individual isoforms exhibiting unique binding specificities. We observe that fru behavioral phenotypes are specificed by either a single, or combination, of isoforms. Finally, we illustrate the utility of these data for the identification of novel sexually dimorphic genoic enhancers, and novel downstream regulators of male-sexual behavior.

Investigation of Drosophila fruitless neurons that express Dpr/DIP cell adhesion molecules

Project description:Drosophila reproductive behaviors are directed by fruitless neurons. A reanalysis of genomic studies shows that genes encoding dpr and DIP Immunoglobulin superfamily (IgSF) members are expressed in fru P1 neurons. We find that each fru P1 and dpr/DIP (fru P1 ∩ dpr/DIP) overlapping expression pattern is similar in both sexes, but there are dimorphisms in neuronal morphology and cell number. Behavioral studies of fru P1 ∩ dpr/DIP perturbation genotypes indicates that the mushroom body functions together with the lateral protocerebral complex to direct courtship behavior. A single-cell RNA-seq analysis of fru P1 neurons shows that many DIPs have high expression in a small set of neurons, whereas the dprs are often expressed in a larger set of neurons at intermediate levels, with a myriad of dpr/DIP expression combinations. Functionally, we find that perturbations of sex hierarchy genes and of DIP-ε change the sex-specific morphologies of fru P1 ∩ DIP-α neurons.

Plantain flour - a beneficial material for the organ and transcriptional profile of kidney of diabetic rats

Project description:This study investigated the ameliorative effects of plantain flour on the organs of diabetic rats, and explored the mechanism from the perspective of kidney transcription. Diabetes was induced in rats by streptozotocin (STZ) and a high-sugar and high-fat diet. The alleviation effects and molecular mechanism of diabetes by plantain flour at different doses (1g/kg·Bw, 2g/kg·Bw, and 4g /kg·Bw) were evaluated based on the analysis of biochemical indicators, histological observations, homeostasis model, and kidney transcriptome. The findings demonstrated that the intervention of plantain flour can reduce the level of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and urea (UREA). The pathological study revealed that plantain flour can alleviate the pathological symptoms of fat accumulation in liver and kidney tissue. According to the homeostasis model, plantain flour was able to reduce insulin resistance in diabetic rats. Transcriptome analysis showed that plantain flour improved dyslipidemia by up-regulating Acsl1, down-regulating Fabp4 and Plin1, and reduced kidney injury by up-regulating Slco1a6, Hsd17b2, Cyp2c24, and Cyp2c11 genes. These results suggested that plantain flour could recover the organ damage caused by diabetes and has the potential to be developed into healthy food.

Identification and functional characterization of a fructose-inducible phosphotransferase system in Azospirillum brasilense Sp7

Project description:Plant growth promoting rhizobacterium Azospirillum brasilense Sp7 utilizes fructose efficiently via a fructose phosphotransferase system (Fru-PTS). Its genome encodes two putative Fru-PTS, each consisting of FruB (EIIA), FruK (Pfk) and FruA (EIIBC) proteins. We compared the proteomes of A. brasilense Sp7 grown with malate or fructose as sole carbon source, and observed upregulation of component proteins of Fru-PTS1 and several proteins involved in the biogenesis of the type 6 secretion system (T6SS) only on fructose.

Project description:L3MBTL2 is a crucial component of ncPRC1.6 and has been implicated in transcriptional repression and chromatin compaction. However, the repression mechanism of L3MBTL2 and its biological functions are largely undefined. CGA encodes the alpha subunit of glycoprotein hormones, however, we showed that CGA plays an individual tumor suppressor role in PDAC. The goals of this part study are to compare transcriptome profiling of PANC-1-shL3MBTL2 cells or PANC-1-CGA-OE cells to their corresponding control cells. By identifying differentially expressed genes (DEGs) and enrichment pathways, the mechanism of L3MBTL2 and CGA in pancreatic cancer can be better studied.

A single-cell transcriptomic atlas of fruitless-expressing neurons in the Drosophila pupal central nervous system

Project description:To examine the repertoires of genes expressed in individual fru P1-expressing neurons, we performed single cell sequencing. The analysis was performed on male and female central nervous system tissues (48-hour pupal stage), from flies that expressed membrane-bound GFP in fru P1 neurons. We chose this developmental stage to gain further insight into how genes direct development of fru P1 neurons, as this is the stage where FruM has peak expression (in ~2,000 neurons).
