RNA-Sequencing analysis of white adipose tissue from wild type mice and Ccdc92 knockout mice

ABSTRACT: Multiple independent genome-wide association studies (GWAS) have uncovered specific genetic variants in the Coiled-Coil Domain Containing 92 (CCDC92) locus that are associated with obesity and insulin resistance in humans. However, the biological function of CCDC92 in metabolism remains unclear. We explored the effects of CCDC92 on obesity and insulin resistance by utilizing wild-type (WT) and Ccdc92 knockout (KO) mice. RNA-seq was used to analyze the gene expression profile regulated by CCDC92 in white adipose tissue.

Insulin resistance rewires the metabolic gene program and glucose utilization in human white adipocytes

Project description:The aim of this study is to define the molecular alterations occurring in human adipose tissue leading to its dysfunction. While obesity (defined by BMI>30) is strongly associated with insulin resistance and metabolic disorders, some individuals with obesity remain insulin sensitive. Therefore, in this study, we have considered a control group (lean, insuln sensitive: BMI<25, HOMA-IR<2), a group with overweight/obesity but without insulin resistance (BMI>25, HOMA-IR<2) and a group with overweight and insulin resistance (BMI>25, HOMA-IR>3) to dissect the transcriptional changes in viscreal adipose tissue due to increased fat mass, and those associated with metabolic disorder.

Project description:The aim of this study was to identify novel long noncoding RNAs (lncRNAs) that are differentially expressed in the subcutaneous region either in obesity or insulin resistance. A number of novel lncRNAs were differentially expressed in the subcutaneous region either in obesity or insulin resistance. Two lncRNAs (termed here as adipocyte specific insulin resistance related lncRNAs (ADLNRIs), ADLNRI 1 and ADLNRI 2) were influenced by both conditions and enriched in adipocytes. Knockdown of either by antisense oligonucleotides in fat cellsadipocytes altered lipolysis and adiponectin secretion, respectively.

A PPARγ-lncRNA axis modulatesthermoneutral remodeling of white adipose tissue

Project description:Interplay between parenchymal energy-storing white adipose cells and thermogenic beige adipocytes contributes to obesity and insulin resistance. Irrespective of cellular origin or specialized niche, adipocytes require the activity of the nuclear receptor peroxisome proliferator activated receptor gamma (PPARγ) for proper function. Exposure to cold or adrenergic signaling enriches thermogenic cells though multiple pathways that act synergistically with PPARγ, however, the molecular mechanisms by which PPARγ licenses white adipose tissue (WAT) to preferentially adopt a thermogenic or white adipose fate in response to dietary cues or thermoneutral conditions are not fully elucidated. Here, we show that a PPARγ-long noncoding RNA (lncRNA) axis integrates canonical and noncanonical thermogenesis to restrain white adipose tissue heat dissipation during thermoneutrality and diet-induced obesity (DIO). Pharmacologic inhibition or genetic deletion of the lncRNA Lexis, enhances UCP-1 dependent and independent thermogenesis. Adipose tissue specific deletion of Lexis counteracted diet-induced obesity, improved insulin sensitivity, and enhanced energy expenditure. Single-nuclei transcriptomics revealed that Lexis regulates a distinct population of thermogenic adipocytes. We systematically map Lexis motif preferences and show that it regulates the thermogenic program through the activity of the metabolic GWAS gene and WNT modulator TCF7L2. Collectively, our studies uncover a new mode of crosstalk between PPARγ and WNT signaling that preserves white adipose tissue plasticity.

Gene expression profiles of white adipose tissues from C57BL/6 mice in the effect of high fat diet

Project description:Metabolic disorders including obesity and insulin resistance have their basis in dysregulated lipid metabolism and low-grade inflammation. In a microarray search of unique lipase-related genes whose expressions are associated with obesity, we found that two secreted phospholipase A2s (sPLA2s), PLA2G5 and PLA2G2E, were robustly induced in adipocytes of obese mice. Analyses of Pla2g5-/- and Pla2g2e-/- mice revealed distinct and previously unrecognized roles of these sPLA2s in diet-induced obesity. PLA2G5 hydrolyzed phosphatidylcholine in fat-overladen low-density lipoprotein to release unsaturated fatty acids, which prevented palmitate-induced M1 macrophage polarization. As such, PLA2G5 tipped the immune balance toward an M2 state, thereby counteracting adipose tissue inflammation, insulin resistance, hyperlipidemia and obesiy. PLA2G2E altered minor lipoprotein phospholipids, phosphatidylserine and phosphatidylethanolamine, and moderately facilitated lipid accumulation in adipose tissue and liver. Collectively, the identification of metabolic sPLA2s adds this gene family to a growing list of lipolytic enzymes that act as metabolic coordinators.
