Experiment Overall Design: PBMCs were treated with 5uL herbal extract for indicated times, followed by RNA isolation with Rneasy kit (Qiagen). Microarray analysis was performed using Affymetrix Human Genome U133 Plus 2.0 Array.

Regulation of inflammatory gene expression in PBMCs by immunostimulatory botanicals.

Project description:Many hundreds of botanicals are used in complementary and alternative medicine for therapeutic use as antimicrobials and immune stimulators. While there exists many centuries of anecdotal evidence and few clinical studies on the activity and efficacy of these botanicals, limited scientific evidence exists on the ability of these botanicals to modulate the immune and inflammatory responses. Using botanogenomics (or herbogenomics), this study provides novel insight into inflammatory genes which are induced in peripheral blood mononuclear cells following treatment with immunomodulatory botanical extracts. These results may suggest putative genes involved in the physiological responses thought to occur following administration of these botanical extracts. Using extracts from immunostimulatory herbs (Astragalus membranaceus, Sambucus cerulea, Andrographis paniculata) and an immunosuppressive herb (Urtica dioica), the data presented supports previous cytokine studies on these herbs as well as identifying additional genes which may be involved in immune cell activation and migration and various inflammatory responses, including wound healing, angiogenesis, and blood pressure modulation. Additionally, we report the presence of lipopolysaccharide in medicinally prepared extracts of these herbs which is theorized to be a natural and active component of the immunostimulatory herbal extracts. The data presented provides a more extensive picture on how these herbs may be mediating their biological effects on the immune and inflammatory responses.

Regulation of microRNA-221, -222, -21 and -27 in articular cartilage subjected to abnormal compressive forces

Project description:Objective: microRNAs (miRs) are small non-coding molecules that regulate post-transcriptional target gene expression and are involved in mechano-regulation of cellular activities in all physiological systems. It is unknown whether such epigenetic mechanisms are regulated in response to increasing magnitudes of load. This study investigated mechano-regulation of miRs in articular cartilage subjected to ‘physiological’ and ‘non-physiological’ compressive loads in vitro as a model system and validated findings in an in vivo model of abnormal joint loading. Design: Bovine full-depth articular cartilage explants were loaded to 2.5MPa (physiological) or 7MPa (non-physiological) (1Hz, 15 minutes) and mechanically-regulated miRs identified using Next Generation Sequencing and verified using quantitative PCR. Downstream targets were verified using miR-specific mimics or inhibitors in conjunction with 3’-UTR luciferase activity assays. Results: A subset of miRs were mechanically-regulated in ex vivo cartilage explants and in vivo joint cartilage. miR-221, miR-222, miR-21-5p, and miR-27a-5p were increased and miR-483 levels decreased with increasing load magnitude. Tissue Inhibitor of Metalloproteinase 3 (TIMP3) and Cytoplasmic Polyadenylation Element Binding Protein 3 (CPEB3) were identified as putative downstream targets. Conclusion: Our data confirms miR-221 and -222 mechano-regulation and demonstrates novel mechano-regulation of miR-21-5p and miR-27a-5p in ex vivo and in vivo cartilage loading models. TIMP3 and CPEB3 are putative miR targets in chondrocytes. Identification of specific miRs that are regulated by increasing load magnitude, and their potential to impact on tissue homeostasis, has direct relevance to other mechano-sensitive physiological systems and provides a mechanism by which load-induced tissue behaviour is regulated, both in health and pathology.

Induction of pluripotency in mouse somatic cells with lineage specifiers (RNA-seq)

Project description:For years, reprogramming factors that induce pluripotency have been identified primarily from ESC-enriched factors, pluripotency-associated factors such as Oct4, Sox2, Klf4, c-Myc, Nanog, Esrrb, Sall4, Utf1, Lin28, PRDM14, and Tet2. Through genome-wide screen we identified novel regulators of reprogramming. Our study is the first proof-of-principle report to suggest a putative general "seesaw model". This model provides novel mechanistic insight into the fundamental understanding of the establishment of cellular identity during programming and reprogramming. Through genome-wide screen to identify novel factors of reprogramming in addition to Oct4, Sox2, Klf4, cMyc.

Case Report: Deep Immune Profiling Links Neutrophil and Plasmablast Dysregulation to Microvascular Damage in post-COVID-19 Multisystem Inflammatory Syndrome of Adults (MIS-A).

Project description:Despite surviving a SARS-CoV-2 infection, some individuals experience an intense post-infectious Multisystem Inflammatory Syndrome (MIS) of uncertain etiology. Children with this syndrome (MIS-C) can experience a Kawasaki-like disease, but mechanisms in adults (MIS-A) are not clearly defined. Here we utilize a deep phenotyping approach to examine immunologic responses in an individual with MIS-A. Results are contextualized to healthy, convalescent, and acute COVID-19 patients. The findings reveal systemic inflammatory changes involving novel neutrophil and B-cell subsets, autoantibodies, complement, and hypercoagulability that are linked to systemic vascular dysfunction. This deep patient profiling generates new mechanistic insight into this rare clinical entity and provides potential insight into other post-infectious syndromes.
