MRNA-seq of worms who raised on control RNAi or prp-19 RNAi were untreated or treated with atp-2 RNAi

ABSTRACT: Mitochondria are the central hub of energy and metabolism of the cell,acting as one of the most important organelles of living cells.Under mitochondrial stresses, cells will trigger mitochondrial unfolded protein response (UPRmt) to maintain mitochondrial homeostasis.We have verified that splicing facotrs are required for the activation of UPRmt.Knocking down of splicing fator prp-19 could supresse UPRmt activation.We analysis the differencial alternative splicing of worms on L4440 RNAi or prp-19 RNAi and untreated or treated with atp-2 RNAi, we found that inhibition of prp-19 interrupts splicing homeostasis of c.elegans.

mRNA-seq of worms and the mitochondrial unfolded protein response (UPRmt)

Project description:To cope with a challenging and unpredictable environment, living systems have evolved several organelle-specific stress responses, e.g. the cytosolic heat shock response (HSR), the endoplasmic reticulum unfolded protein response (UPRER) and the mitochondrial unfolded protein response (UPRmt). UPRmt monitors mitochondrial function and homeostasis in general. However, the mechanism of UPRmt remains largely unexplored. Here we identified that histone deacetylase HDA-1 is associated with homeobox domain-containing protein DVE-1 in UPRmt activation in Caenorhabditis elegans. Knocking down ATP synthase subunit atp-2 generates mitochondrial stress and induces UPRmt. After analyzing the mRNA profiles of worms on L4440 RNAi, hda-1 RNAi or dve-1 RNAi and untreated or treated with atp-2 RNAi, we found that 283 hda-1_dependent genes and 218 dve-1_dependent genes were upregulated in response to atp-2 RNAi.

Transcriptome analysis of zcIs13 and zcIs13; Punc-119_ds-fzo-1 RNAi in Caenorhabditis elegans

Project description:Mitochondrial functions across different tissues are regulated in a coordinated fashion to optimize fitness of the organism. Mitochondrial unfolded protein response (UPRmt) can be non-autonomously elicited by mitochondrial perturbation in neurons, but neuronal signals that propagate such response and its physiological significance remains incompletely understood. Here we show that in C. elegans, loss of neuronal fzo-1/Mitofusin induces non-autonomous UPRmt through multiple neurotransmitters and neurohormones, including acetylcholine, serotonin, glutamate, tyramine and insulin-like peptides. Neuronal fzo-1 depletion also triggers non-autonomous mitochondrial fragmentation, which requires autophagy and mitophagy genes. Systemic activation of UPRmt and mitochondrial fragmentation in C. elegans via perturbing neuronal mitochondrial dynamics improves resistance to pathogenic Pseudomonas infection, which is supported by transcriptomic signatures of immunity and stress response genes. We propose that C. elegans surveils neuronal mitochondrial dynamics that coordinate systemic UPRmt and mitochondrial connectivity for pathogen defense and optimized survival under bacterial infection.

miRNA sequencing detecting miRNA cargo from PRP derived exosomes isolated from young donors and old donors

Project description:We detected and compared the miRNAs contained in Y-PRP-exos or O-PRP-exos. The results showed that most miRNAs (38) can be identified in both Y-PRP-exos and O-PRP-exos. In addition, hsa-let-7f-5p, hsa-miR-16-5p and hsa-miR-126-3p were top 3 enriched miRNAs in PRP-exos of all donors. These results indicated the heterogeneity of miRNAs in PRP-exos was small, regardless of age. However, the abundance of miRNAs in Y-PRP-exos and O-PRP-exos was quite different, especially in high-ranking miRNAs. The expression of hsa-miR-16-5p and has-let-7f-5p in O-PRP-exos in two old donors was only approximately 1/4-1/2 of that in Y-PRP-exos in two young donors
