The profiles of mRNAs, lncRNAs and circRNAs in browning of human adipose derived stem cells

ABSTRACT: To investigate the profiles of mRNAs, lncRNAs and circRNAs in browning of human adipose derived stem cells (hADSCs), we obtained hADSCs from omentum adipose tissues of 5 patients on who were performed abdominal operation and derived them into brown adipocytes. Then we employed microarray profiling the expression of lncRNAs, circRNAs and mRNA of hADSCs and their derived brown adipocytes.

We found robust browning of inguinal white adipose tissue in UCP1/ApoE-DKO mice, but not in ApoE-KO mice, under high-fat diet condition. We used microarray to determine the genes specifically regulated in the browning white adipose tissue in UCP1/ApoE-DKO mice.

BAF60a deficiency uncouples chromatin accessibility and cold sensitivity from white fat browning

Project description:Brown and beige fat share a remarkably similar transcriptional program that supports fuel oxidation and thermogenesis. The chromatin-remodeling machinery that governs genome accessibility and renders adipocytes poised for thermogenic activation remains elusive. Here we found that BAF60a, a subunit of the SWI/SNF chromatin-remodeling complexes, serves an indispensible role in cold-induced thermogenesis in brown fat. BAF60a maintains chromatin accessibility for key thermogenic genes in close proximity to PPARg and EBF2 binding sites. Surprisingly, fat-specific BAF60a inactivation triggers more pronounced browning of inguinal white adipose tissue that is linked to induction of MC2R, a receptor for the pituitary hormone ACTH. Elevated MC2R expression sensitizes adipocytes and BAF60a-deficient adipose tissue to thermogenic activation in response to ACTH stimulation. These observations reveal an unexpected dichotomous role of BAF60a-mediated chromatin remodeling in transcriptional control of brown and beige gene programs and illustrate a pituitary-adipose signaling axis in the control of thermogenesis.

Knocking down CDKN2A in 3D hiPSC-derived brown adipose progenitors potentiates differentiation, oxydative metabolism and browning process

Project description:Human induced pluripotent stem cells (hiPSCs) have the potential to be differentiated into any cell type, making these cells a relevant tool for therapeutic purposes such as cell-based therapies. In particular, they show great promise for obesity treatment as they represent an unlimited source of brown/brite adipose progenitors (hiPSC-BAPs), although the low brown/brite adipocyte differen-tiation potential in 2D culture represents a strong limitation for clinical use. In adipose tissue, be-sides to its cell cycle regulator functions, the cyclin-dependent kinase inhibitor 2A (CDKN2A) lo-cus modulates the commitment of stem cells to the brown-like type fate, mature adipocyte energy metabolism and browning of adipose tissue. Here, using a new method of hiPSC-BAPs 3D culture, via the formation of an organoid-like structure, we silenced CDKN2A expression during hiPSC-BAP adipogenic differentiation and observed that knocking down CDKN2A potentiates adipogenesis, oxidative metabolism and browning process, resulting in brown-like adipocytes by promoting UCP1 expression and beiging markers. Our results suggest that modulating CDKN2A levels could be relevant for hiPSC-BAPs cell-based therapies.

Browning of human adipocytes requires KLF11 and reprogramming of PPARγ super-enhancers

Project description:Here we have characterized the transcriptional processes underlying the formation of human brown in white (i.e. brite) adipocytes using a genome-wide approach. We show that the browning process is associated with reprogramming of peroxisome proliferator-activated receptor γ (PPARγ) binding to form brite adipocyte-selective PPARγ super-enhancers that appear to play a key role in activation of brite adipocyte-selective genes. We identify the KLF11 gene based on its association with a PPARγ super-enhancer and show that KLF11 is a novel browning factor directly induced by rosiglitazone and required for the activation of brite adipocyte-selective gene program by rosiglitazone. Transcriptomics to discover splice variants involved in the browning of adipose tissue

Project description:Adipose tissue is the major depot for energy storage. Recent studies have shown that at least three types of adipocytes can be distinguished depending on their anatomical locations : 1) The classic brown adipocytes, i.e., brown adipose tissue (BAT); 2) The 'brite' (brown-in-white) adipocytes, i.e. inguinal white adipose tissue (iWAT); 3) The 'true' white adipocytes, i.e., epididymal white adipose tissue (eWAT). Two strains of mice (SV129 and C57BL/6J) were used in this study. SV strain is resistant to obesity and latter is prone to obesity. Pre-adipocyte cells were isolated from subcutaneous tissue (iWAT) to create four groups of cell cultures per strain of mouse.

Gene expression signature of brown and inguinal white fat of mice kept at 30°C vs. 5°C

Project description:Activation and recruitment of thermogenic cells in human white adipose tissues (“browning”) can counteract obesity and associated metabolic disorders. However, quantifying the effects of therapeutic interventions on browning remains enigmatic. Here, we devise a computational approach, profiling of fat tissue types (ProFAT), for the quantification of thermogenic potential of heterogeneous fat biopsies based on the prediction of white and brown adipocytes content from raw gene expression profiles. ProFat systematically integrates 103 mouse fat-derived transcriptomes to identify unbiased and robust gene signatures of brown and white adipocytes. Application of ProFAT to 80 mouse and 97 human transcriptional profiles from 14 independent studies correctly predicts browning capacity upon various physiological and pharmacological stimuli. Our study represents the most exhaustive comparative analysis of public data on adipose biology towards quantification of browning after personalized medical intervention. ProFat is freely available and should become increasingly powerful with the growing wealth of transcriptomics data.
