Project description:Aedes aegypti mosquitoes are a persistent human foe, transmitting arboviruses including dengue when they feed on human blood. Mosquitoes are intensely attracted to body odor and carbon dioxide, which they detect using ionotropic chemosensory receptors encoded by three large multi-gene families. Genetic mutations that disrupt the olfactory system have modest effects on human attraction, suggesting redundancy in odor coding. The canonical view is that olfactory sensory neurons each express a single chemosensory receptor that defines its ligand selectivity. We discovered that Ae. aegypti uses a different organizational principle, with many neurons co-expressing multiple chemosensory receptor genes. In vivo electrophysiology demonstrates that the broad ligand-sensitivity of mosquito olfactory neurons depends on this non-canonical co-expression. The redundancy afforded by an olfactory system in which neurons co-express multiple chemosensory receptors may increase the robustness of the mosquito olfactory system and explain our long-standing inability to disrupt the detection of humans by mosquitoes.

Single nucleus RNA-Seq is not suitable for detection of microglial activation genes in humans

Project description:Single nucleus RNA-Seq (snRNA-Seq) is used as an alternative to single cell RNA-Seq, as it allows transcriptomic profiling of frozen tissue. However, it is unclear whether snRNA-Seq is able to detect cellular state in human tissue. Indeed, snRNA-Seq analyses of human brain samples have failed to detect a consistent microglial activation signature in Alzheimer’s Disease. Our comparison of microglia from single cells and single nuclei of four human subjects revealed that, while the majority of genes showed similar relative abundances in cells and nuclei, a small population of genes (~1%) was depleted in nuclei compared to whole cells. This population was enriched for genes previously implicated in microglial activation, including APOE, CST3, SPP1, and CD74, comprising 18% of previously-identified microglial disease-associated genes. Given the low sensitivity of snRNA-Seq to detect many activation genes, we conclude that snRNA-Seq is not suited to detecting cellular activation in microglia in human disease.

Comparative transcriptomics of chemosensory tissues between Drosophila species

Project description:These data contain RNA-seq samples generated from the main chemosensory organs of closely related Drosophila species (D. melanogaster, D. sechellia, D. simulans, D. santomea, D. erecta, and D. suzukii) from larava (head) and adults (antenna, forelegs, proboscis with maxillary palps, ovipositors (female only) for both males and females. The purpose for generating these data was to carry out evolutionary analyses of gene expression differences between the six species.

snRNA-seq of whole hearts from adult BL6 mice

Project description:We used snRNA-seq to investigate an entire adult mammalian heart of BL6 mice. Whole hearts were harvested from 4 male mice (12 weeks) after cervical dislocation. The hearts were pooled and nuclei isolated using the Nuclei PURE Prep isolation kit (Sigma-Aldrich, Darmstadt, Germany) according to the manufacturer’s protocol. Sequencing was conducted by Genewiz (Leipzig, Germany) on the 10xGenomics system. Single nuclei were captured in droplet emulsions and snRNA-seq libraries were constructed as per the 10x Genomics protocol using GemCode Single-Cell 3′ Gel Bead and Library V3 Kit. RNA was controlled for sufficient quality on an Agilent 2100 Bioanalyzer system and quantified using a Qubit Fluorometer.

The contribution of temporal coding to odor coding and odor perception in humans.

Project description:Whether neurons encode information through their spike rates, their activity times or both is an ongoing debate in systems neuroscience. Here, we tested whether humans can discriminate between a pair of temporal odor mixtures (TOMs) composed of the same two components delivered in rapid succession in either one temporal order or its reverse. These TOMs presumably activate the same olfactory neurons but at different times and thus differ mainly in the time of neuron activation. We found that most participants could hardly discriminate between TOMs, although they easily discriminated between a TOM and one of its components. By contrast, participants succeeded in discriminating between the TOMs when they were notified of their successive nature in advance. We thus suggest that the time of glomerulus activation can be exploited to extract odor-related information, although it does not change the odor perception substantially, as should be expected from an odor code per se.
