Proteomic profiling of two commonly used laboratory yeast strains: W303 and BY4742

Project description:The yeast Saccharomyces cerevisiae is a powerful model system that is often used to expand our understanding of cellular processes and biological functions. Although many genetically well-characterized laboratory strains of S. cerevisiae are available, they may have different genetic backgrounds which can confound data interpretation. Here, we profile and compare the proteomes of two common laboratory yeast background strains, W303 and BY4742, in both exponential and stationary growth phases using isobaric tag-based mass spectrometry. We quantified over 4,400 proteins, hundreds of which showed differences in abundance between strains and/or growth phases. Moreover, we used proteome-wide abundance to profile the mating type of the strains used in the experiment, the auxotrophic markers and associated metabolic pathways, as well as to investigate differences in particular classes of proteins, such as the pleiotropic drug resistance (PDR) proteins. This study is a valuable resource that offers insight into mechanistic differences between two common yeast background strains and can be used as a guide to select a background that is best suited for addressing a particular biological question.

Metabolically engineered urea degrading and urea importing Sake yeast strains K7 (WT), K7 Dur1,2 and K7 Dur3

Project description:Excess/residual urea is a pervasion problem in wine and Sake fermentation. We sought to reduce residual urea levels (to reduce ethyl carbamate leves) by engineering the Sake yeast strain K7 to constitutively express either the urea amidolyase (Dur1,2) or urea importer (Dur3). We sought to then compare the gene expression profiles of the metabolically engineered yeast strains to the parental strain during fermentation. Engineered strains would hopefully have gene expression profiles that were minimally different from the parental strain.

Project description:Furfural, phenol and acetic acid, generated during the cellulosic material pretreatment, are the representative inhbitors to yeast used for ethanol production. The responses to multi-inhbitors in industrial yeast and the tolerant strain were analyzed. We analyzed the transcriptome of the parental and tolerant strains in the presence of multi-inhibitors. Parental and tolerant strains were collected at the exponential stage in the presence of multi-inhibitors. The reference samples for industrial yeast and adapted strains were collected at the same growth stage in the absence of inhibitors.
