Single-Cell Transcriptome Profiling Reveals Intratumoral Heterogeneity and Molecular Features in Ductal Carcinoma In Situ of the Breast

ABSTRACT: This sample was collected to conduct spacial transcriptome analysis of breast cancer. We analyzed heterogeneity of HER2+ ducal cacinoma in situ (DCIS). We obtained transcriptome data from 1,521 distinct spots. By an unsupervised dimensionality reduction approach through UMAP, the data were decomposed into 5 clusters (Clusters 0 to 4). The 2 luminal epithelial clusters were identified, however ERBB2/HER2 expression level was different in these clusters. Unsupervised clustering analysis showed clear separation of the cells from the 2 clusters. Analysis of canonical pathways and upstream regulators by IPA indicated functionally distinct luminal epithelial populations in one HER2+ DCIS sample.
