Compare the knock-down effects of SLC2A11-MIF and two parent genes in Hela cell line

Project description:Total RNA were extracted from five knocking down treated Hela cells using siRNA targeting fusion gene SLCC2A11-MIF and parental genes To investigate the function of fusion gene and parental gene, we designed siRNAs specifically knocking down the fusion RNA SLC2A11-MIF and parental gene SLC2A11 respectively. Totally we have five samples. One sample is control, two siRNAs target parental genes and other two siRNAs target fusion genes. After 48hrs siRNA treatment, the RNAs were extracted from cells, and followed by RNA microarray.

MAGT1 is involved in HeLa cell proliferation through regulating p21 expression, S-phase progress and ERK/p38 MAPK MYC axis

Project description:Background: Magnesium transporter subtype 1 (MAGT1) is involved in animal development and cell differentiation. Thus far, researches on MAGT1 are mainly focused on Cardiomyocyte regulation and differentiation, but rare research has been reported on cell proliferation, especially concerning the underlying molecular mechanisms. Methods: HeLa cells were transiently knock down with different SiRNAs and processed for RNA sequencing and bioinformatics analysis, western blot, real-time PCR as well as cell cycle assays. Results: We showed that cell proliferation was significantly restricted with S-phase arrest in MAGT1 knock down cells, which was further confirmed by the up expression of p21, CCNA1 and CCNB1, while down expression of CCND1, CCNE1 and CDK4. Knocking down MAGT1 also resulted in significant transcriptional expression changes of 1599 target genes analyzed by RNA sequencing. The bioinformatics analysis showed that MAGT1 was related to MAPK signaling pathway. Western blot analysis confirmed that the phosphorylation of ERK1/2 and p38 exhibited a remarkable reduction in MAGT1 downregulated group. Conclusion: MAGT1 is critical in HeLa cell proliferation, S-phase progress and ERK/p38 MAPK signaling pathway, indicating that MAGT1 might be a novel target of anticancer research.

Comparison of PPIH knockdown in HeLa cells compared to scrambled shRNA control

Project description:HeLa cells transduced with shRNA from MISSION library (TRCN0000289677) using lentiviral delivery system. HeLa cells transduced with scrambled shRNA, gifted from Dr. Mauricio Reginato. Sequence is 5′-CCTAAGGTTAAGTCGCCCTCGCTCTAGCGAGGGCGACTTAACCTT-3′ Peptidyl-prolyl isomerase H (PPIH) is a cyclophilin, an enzyme that interconverts cis and trans isomers of proline. PPIH associates with the human spliceosome, the complex and dynamic machinery that removes intronic sequence from pre-messenger RNA (pre-mRNA). Although PPIH affects the ability of the spliceosome to assemble and catalyze splicing in vitro, little is known about what PPIH does to splicing in vivo. To understand the function of PPIH in the nucleus, we knocked down PPIH in human cells. We characterized a set of alternative splicing and transcriptional events that are PPIH-responsive. We used these splicing and transcriptional bioassays to show first that PPIH-responsive events are largely specific, even within the cyclophilin family. We then identified a role for proline isomerization in transcriptional regulation, but not in regulation of alternative splicing. The development of a bioassay for PPIH function can be used to answer fundamental questions about the role of spliceosomal proteins in regulating splicing and other nuclear functions.
