Project description:Abdominal subcutaneous adipose tissue was obtained from obese individuals before, after an 8-week low calorie diet and after an ad libitum 26-week weight control phase. Clinical investigations including subcutaneous adipose tissue biopsies were performed at baseline and at the end of each phase. Main clinical outcome measures were body weight, body composition, energy intake, metabolic biochemistry. Adipose tissue gene expression profiling and lipidomics from the same samples were also assessed.

Project description:We established a useful protocol for generating insulin -producing cells (IPCs) from canine adipose mesenchymal stem cells (cAD-MSCs). IPCs were induced from cAD-MSCs with the three-stepwise protocol. For the first stage , cAD-MSCs was induced to definitive endoderm (DE) with the cooperation of Chir99021 and Activin A for 3 days. For the second stage, pancreatic endocrine (PE) progenitor was induced from DE after treatment with taurine, retinoic acid, FGF2 EGF, TGFβ inhibitor, dorsomorphin, nicotinamide, and DAPT for the next 5 days. In the last stage, the combination of taurine, nicotinamide, Glp-1, forskolin, PI3K inhibitor, and TGFβ inhibitor was used to yield efficiently functional IPCs from PE precursors for 5 days. To indentify differences in the expression pattern of genes related to pancreatic beta cell development, we performed real time quantitative polymerase chain reaction (RT-qPCR).

Project description:Adult female mice (C57BL/6) were super-ovulated by intraperitoneal hormone injections with the interval of 48 hours between injections. The hormones administered were PMSG followed by hCG (5IU/0.1ml, Sigma-Aldrich). Following the second hormonal treatment, the females were mated with males (1:1, C57BL/6). Zygotes, 2-cell and 4-cell and blastocysts were collected 26, 48 and 52 hours post hCG. Prior to cell collection, the zona pelucida was removed by Tyrode s acid solution. The 2- and 4-cell embryos were then immersed in Trypsin solution (Invitrogen) with RNase inhibitor (1 IU/ul, Clontech) and BSA (50 ug/ul) for the separation of the blastomeres. The separated blastomeres were snap frozen, and maintained at -80 C until processed. Twelve zygotes, 20 2-cell and 9 4-cell embryos were collected for single cell qPCR. In addition, we prepared with a pool of 10 zygotes and a pool of 50 zygotes as positive controls. The cells and pools were subjected to reverse transcription with SuperScript VILO cDNA Synthesis kit (Life Technologies) as recommended by the manufacturer. DELTAgene Assays were custom designed for 94 genes associated with embryonic development or regulation of gene expression, as well as 2 housekeeping genes. The cDNA from cells and pools were subjected to target specific amplification for 20 cycles. Pre-amplified template was diluted and subjected to primer specific amplification with SooFast EvaGreen Supermix (Biorad) on 96.96 array chip and BiomarkHD System (Fluidigm). Project description:ObjectivesThis study describes parasite kinetics in the blood of visceral leishmaniasis patients treated with liposomal amphotericin B (L-AmB) or a preformed fat emulsion of amphotericin B (ApL) using real-time quantitative PCR (qPCR).MethodsForty-six patients were treated with a single dose (15 mg/kg of body weight) of either L-AmB (n = 13) or ApL (n = 33). qPCR was used to estimate parasite kinetics by detection of Leishmania donovani DNA using kinetoplast DNA-specific primers in peripheral blood samples using an absolute quantification method.ResultsThe mean parasite load decreased from baseline (day 0) values of 894.07 and 980.48 to 71.72 and 211.52 parasite genomes/mL at day 7 in L-AmB and ApL groups, respectively, and at day 30 these further declined to 8.30 and 133.98 parasite genomes/mL, respectively. At day 30 post-treatment evaluation, the decline in parasite load was significantly greater (P = 0.024) with L-AmB compared with ApL. Four of 33 patients in the ApL group failed treatment (1 primary failure and 3 relapses) with the presence of parasites, whereas all patients in the L-AmB group were cured at 6 month follow-up.ConclusionsqPCR can be a tool to measure parasite dynamics accurately and provide a marker to measure the efficacy of various drugs. It can be used as a test of cure, allowing us to do away with invasive and risky methods such as splenic or bone marrow aspiration.
