Study of Gastric Cancer

ABSTRACT: The objective of my study was to identify the genes differentially expressed in gastric adenocarcinoma associated with prevalent risk factor such as tobacco use (Meiziol, Tuibur etc) in high-risk Indian population. The Tumor tissue and matched normal tissue distant to the tumor were collected during endoscopy in RNA later, Snap-frozen in liquid nitrogen and stored at -70°C until processed. Data of clinicopathologic parameters were obtained from patients’ clinical report, operative notes and pathologic report. Institutional Human ethics committee approved the study.

Project description:The objective of this study was to identify the genes differentially expressed in non small cell lung carcinoma associated with prevalent risk factor such as smoking and betel quid chewing in high-risk north eastern Indian population. The tumor biopsies and matched normal tissue from distant site were collected in RNA later, snap-frozen in liquid nitrogen and stored at -70°C until processed. Data of clinicopathologic parameters were obtained from patients’ clinical and pathologic report. Institutional human ethics committee approved the study. Total RNA was isolated from tumor biopsies and matched normal of five patients. Total RNA from normal tissue from three patients was pooled in one slide and normal tissue from another two patients was pooled in another slide. Individual slide were used for each tumor samples. Proteomic profiling comparison for plasma from patients with acute ischemic stroke under rt-PA treatment

Project description:In this study, a total of 10 patients who received rt-PA were enrolled. Plasma samples were collected before and 1 hour after the rt-PA infusion for proteomic analysis. Blood sample collection has been approved by Tianjin Medical University General Hospital Ethics Committee, and collection was conducted according to the Declaration of Helsinki. Informed consent was obtained from all participants, and all experiments were conducted in accordance with Chinese laws and institutional guidelines.

Project description:In this study, neutrophils isolated from human peripheral blood were cultured with PBS, tPA(10μg/ml) or tPA+HRG (0.5μm) for 2.5 hours at 37 ° C . Then, neutrophils were collected for proteomic analysis (n=6). Gene expression profiling of cervical cancer tissues at progressive FIGO defined stages

Project description:This is a study indented to investigate the alterations in the molecular profile of cervix uteri, as it progresses thorough various FIGO stages of carcinoma, by means of global gene expression profiling, in a cohort of Indian patients. In addition expresion profiles of early and advanced stage cervical cancer were compared to identify biomarkers and therapeutic targets for the advanced stages. Cervical cancer and non-malignant cervical tissues are obtained from consenting patients following hospital ethics committee approved protocols. The samples are profiled against universal Human reference RNA from Stratagene on 19K EST microarrays from Microarray Center, University Health Network, Toronto, Canada. Biological replicates: Normal = 4; Cervical cancer Stage I = 8; Cervical cancer Stage II = 9; Cervical cancer Stage III = 8. One replicate per array.

RNA difference in superficial temporal artery between moyamoya disease and normal control

Project description:We collected the Superficial temporal artery (STA) tissues from patients with Moyamoya disease who underwent combined direct and indirect bypass surgery and patients with brain trauma requiring craniotomy in the Department of Neurosurgery, First Affiliated Hospital of USTC (Anhui Provincial Hospital). One part was fixed in 10% neutral formalin solution, and the other part was stored in a refrigerator at -80 ℃. All protocols using human specimens were approved by the ethics committee of the First Affiliated Hospital of USTC (Anhui Provincial Hospital). Written informed consent was obtained from all patients. All protocols were approved by the Institutional Review Board of the First Affiliated Hospital of USTC (Anhui Provincial Hospital).Total RNA was extracted from the STA tissues using Trizol reagent (Invitrogen, Carlsbad, CA, USA) according to the manufacturer's protocol. The integrity and concentration of RNA were detected using an Agilent Bioanalyzer 2100 system (Agilent Technologies, Santa Clara, Calif., USA), enriched and purified with Oligo (dT) -bearing magnetic beads. RNA sequencing was performed by Anoroad (Beijing, China).

Characterization of plasma proteins of HIV/AIDS patients underfirst line anti-retroviral therapy

Project description:The retrospective cohort study was focused on the first line ART patients who were enrolled in the antiretroviral therapy center, Sarojini Naidu Medical College, Agra, India from December 2009 to November 2016. The main criteria for inclusion was based on the immunological marker, i.e. CD4+ count (<350). The all age group adults and adolescents were included in this study. The informed consents were obtained from all patients. A patient information leaflet was used for collection of sociodemographic and clinical profile data. This study protocol was approved by Institute human ethics committee. The Institute ethics committee is constituted as per the guidelines directed by Indian Council of MedicalResearch. Ten ml of blood samples were collected from a total of six AIDS patients after counselling whose CD4+ count was <350 cells/µl. Then plasma was separated by centrifugation at 2000g for 10 minutes. Then plasma was stored at -800C for estimation of viral load and genotyping was carried out at NIRT, ICMR, Chennai, Tamilnadu, India.The viral load was estimated using Abbott automated m2000rt instrument. Those who were having>1000 copies/ml called as a drug resistant and < 1000 copies/ml called as drug responder. Among them, 3 patients were having high viral load >1000 copies/ml (drug resistant and 3 patients were with <1000 copies/ml (drug responder). The 3 treatment failure (>1000 copies/ml) samples were further considered for genotype analysis. Highly abundant proteins (Albumin and IgG) were depleted using the Aurum–Serum protein mini kit (Biorad, USA). The depleted plasma samples were analyzed in SWATH-MS. Then, the proteins were identified and characterized using the SWATH analysis.
