Characterising the chromatin dynamics upon HER2 overexpression

ABSTRACT: We performed ATAC-seq to cells isolated from 3D cell culture to understand their 3D conformational change. An indcuible HER2 expression was induced from 0-48 hours and their ATAC-seq performed to their acini. This allowed us to isolate their chomatin changes at the very earliest time-points, which has not been performed to date.

Project description:HER2 transduced cells which we will refer to as HER2-DOX –. These cells are 99% GFP positive (i.e., 99% cells have HER2 transduced, un-induced). As a control, we had GFP empty vector transduced MCF10A cells (95% have GFP transduced). Both cell types in triplicates. We had 4 time-points 0h, 30 mins, 4hours, and 7 hours (time duration for which HER2 will be induced). DOX was added to the GFP-MCF10A cells as a control. Only 1ug/ml of DOX was be used.

Transcription profiling by array of human stromal cells cultured with or without 3D BPH-1 prostate epithelial cells

Project description:BPH-1 prostate epithelial cells when cultured in Matrigel form 3D acini which closely recapitulate the tissue architecture of the prostate. The presence of primary stroma increases epithelial adhesions and causes changes in morphology. To identify the stromal signals which control actin/cadherin dynamics in epithelial cells we performed a microarray analysis on the stroma using Affymetrix U133 plus 2.0 arrays. Primary stroma from 7 different patients were cultured with (PSE) or without (PS) 3D BPH-1 epithelial acini to determine the gene expression changes within the stroma. The RNA from the Primary epithelia and stromal cultures were used at passages 1 or 1-3, respectively.

PMCA2 Regulates HER2 Expression, Cell-Surface Localization and Signaling, and Promotes HER2- Mediated Breast Cancer

Project description:In the normal breast, PMCA2 transports calcium into milk. It is also upregulated in breast cancer cells and high tumor levels predict increased mortality in patients. In this study, we examined interactions between PMCA2 and HER2. We find that PMCA2 and a scaffolding protein, NHERF1, interact together with HER2 in specific membrane domains protruding from the surface of breast cancer cells. Knocking down PMCA2 or NHERF1 increases intracellular calcium and leads to the ubiquitination, endocytosis and degradation of HER2 after receptor activation. This is associated with reductions in HER2 expression and signaling. Manipulating PMCA2 levels regulates proliferation and apoptosis of breast cancer cells in vitro and knocking out PMCA2 dramatically inhibits the formation of hyperplasia and tumors in MMTV-Neu mice. Gene expression profiling was performed on control SKBR3 cells vs. PMCA2, NHERFR1 and HER2 knock down cells. Each group has 6 duplicates.

Study of gene targets regulated by oncogenic PIK3CA and/or Her2

Project description:Mutant PIK3CA and Her2 genes are oncogenic and their co-existence in breast cancer has been well identified. However, the gene targets and cell signalling pathway regulated by mutant PIK3CA, Her2 and both of PIK3CA and Her2 have not been well studied. We established stable cell models through transfecting mutant PIK3CA, Her2 and both mutant PIK3CA and Her2 into MCF10A cells and performed Affymetrix microarray to identify downstream target genes controlled by either mutant PIK3CA, Her2 or both PIK3CA and Her2.

Role of HER2 in breast cancer stem cells

Project description:HER2 overexpression results in expansion of breast cancer stem cells (CSCs). However, the underlying mechanism remains elusive. We performed microarray analysis to obtain HER2-induced gene expression profiles in MCF10A breast epithelial cells and found that an array of inflammatory cytokine genes, including IL-1 and IL-6, are activated. Our study indicates that targeting the HER2-IL-1α-IL-6 axis provides an effective approach for eliminating of CSCs and treating HER2-positive breast cancer.
