Next generation sequencing of transcripts associated to four selected RNA-binding proteins in Tripanosoma brucei

ABSTRACT: We report the sequencing of mRNAs associated with four Trypanosoma brucei translation intiation proteins EIF4E3, EIF4E4, EIF4G3 and EIF4G4. To investigate if the proteins have redundant functions, we analyzed mRNAs subpopulations in cells submitted to RNAi of EIF4E3 and EIF4E4. The reduction of EIF4E3 protein levels did not change the profile of mRNA populations associated to EIF4E4.

Project description:Gene expression in pathogenic protozoans of the family Trypanosomatidae has several novel features, for example in protein synthesis there are multiple eIF4F-like complexes. The eukaryotic eIF4F complex, formed by the eIF4E, eIF4G and eIF4A subunits, is responsible for the canonical selection of mRNAs required for the initiation of mRNA translation. The best-known complexes implicated in translation in trypanosomatids are based on two related pairs of eIF4E and eIF4G subunits (EIF4E3/EIF4G4 and EIF4E4/EIF4G3), whose functional distinctions have yet to be properly identified. Here, to define the interactome associated with both complexes in Trypanosoma brucei procyclic forms, we performed parallel immunoprecipitation experiments followed by identification of proteins co-precipitated with the two sets of tagged eIF4E and eIF4G subunits. A number of different protein partners, including RBPs and RNA helicases, were found to specifically co-precipitate with each complex. Highlights with the EIF4E4/EIF4G3 pair include RBP23, PABP1, EIF4AI and the CRK1 kinase. Co-precipitated partners with the EIF4E3/EIF4G4 pair are more diverse and include DRBD2, PABP2 and different zinc-finger proteins and RNA helicases.EIF4E3/EIF4G4 are known to be essential for viability and to better define their role.

In vitro generation of human high-density-lipoprotein-resistant Trypanosoma brucei brucei

Project description:The host range of African trypanosomes is influenced by innate protective molecules in the blood of primates. A subfraction of human high-density lipoprotein (HDL) containing apolipoprotein A-I, apolipoprotein L-I, and haptoglobin-related protein is toxic to Trypanosoma brucei brucei but not the human sleeping sickness parasite Trypanosoma brucei rhodesiense. It is thought that T. b. rhodesiense evolved from a T. b. brucei-like ancestor and expresses a defense protein that ablates the antitrypanosomal activity of human HDL. To directly investigate this possibility, we developed an in vitro selection to generate human HDL-resistant T. b. brucei. Here we show that conversion of T. b. brucei from human HDL sensitive to resistant correlates with changes in the expression of the variant surface glycoprotein (VSG) and abolished uptake of the cytotoxic human HDLs. Complete transcriptome analysis of the HDL-susceptible and -resistant trypanosomes confirmed that VSG switching had occurred but failed to reveal the expression of other genes specifically associated with human HDL resistance, including the serum resistance-associated gene (SRA) of T. b. rhodesiense. In addition, we found that while the original active expression site was still utilized, expression of three expression site-associated genes (ESAG) was altered in the HDL-resistant trypanosomes. These findings demonstrate that resistance to human HDLs can be acquired by T. b. brucei. Keywords: Trypanosoma, VSG, antigenic switching, HDL-resistance
