A Bidirectional Single-Cell Migration and Retrieval Chip for Quantitative Study of Dendritic Cell Migration

ABSTRACT: We describe a microfluidics-based single-cell migration platform that enables high-throughput and precise bidirectional chemotaxis assays (BM-Chip), which allows controllable retrieval of cell subpopulations that have different migratory potentials. We investigated dendritic cell (DC) migration in response to different chemoattractants and inhibitors. We quantitatively described DC migration patterns and retrieved DC subpopulations of different migratory potentials for differential gene expression analysis.
