AAV-mediated LRG1 overexpression in diabetic mice

ABSTRACT: To investigate the effect of LRG1 overexpression on gene expression of eWAT from db/db mice, we transduced 4-week-old db/db mice with AAV8-eGFP or AAV8-LRG1-FL and harvested eWAT at 7 weeks and 10 weeks of age. We then performed gene expression profiling analysis using data obtained from RNA-seq of eWAT from eGFP- and LRG1-overexpressing mice at two time points.

A truncated reverse transcriptase enhances prime editing by split AAV vectors

Project description:To investigate if the truncated PE can be dilivered by dual AAV8 vectors for in vivo prime editing. We injected the dual AAV8 into 10-week-old C57BL/6J mice . Livers were isolated 4 weeks after injection and next generation sequencing showed an average of 1.4% and 5.4% precise prime editing with the low and high AAV doses, respectively (Figure 4D ). This demonstrates that PECO-Mini can be efficiently delivered by dual AAVs for in vivo prime editing.

Benchmarking AAV-Cre mouse models for RNA-seq

Project description:This study reports the baseline effects of systemic (i.v.) administration of AAV8-TBG-null and AAV8-TBG-Cre on the liver of male, wild-type C57Black6/J mice. We performed polyA RNAseq of whole liver lysates from mice 2, 4 and 7 days post AAV8-TBG-null or AAV8-TBG-Cre administration and compared their transcriptome with that of male mice of similar age that didn't receive any AAV8 (day 0). We report some changes in gene expression of genes related to circadian rythm and response to infection at certain timepoints. Our results demonstrate that AAV8-TBG vectors can have minimal effects on the murine liver transcriptome, something that needs to be considered and controlled when using this system for mouse experiments. Finally, we hope that this dataset will help other researchers to correctly interpret data obtained from mouse experimentation using AAV8-TBG vectors.

Previous studies showed that it has great importance in epithelial–mesenchymal transition in colorectal cancer and could promote glioma cell lines invasion and migration. Despite the intimate relationships between LRG1 and migration in cancer research, its role in wound repair or re-epithelialization has not been uncovered. So we dicided using keratinocytes to see its transcriptomes changing after LRG1 addition.

Phospho-antibody microarray anayses for control and DG-LRG1 treated HUVEC cells.

Project description:Deglycosylated-leucine-rich α-2-glycoprotein1 (DG-LRG1) as well as LRG1 was discovered to promote angiogenesis under diabetes mellitus condition through TGF-β independent binding to endoglin. To examine the signaling pathways triggered by DG-LRG1, we subjected whole-cell protein lysates of control and DG-LRG1 treated HUVECs to a Phospho Explorer antibody array analysis using commercial antibody array assay kit (Full Moon Biosystems, Inc.). Samples were probed against 1318 site-specific and phospho-specific antibodies with two replicates per antibody printed on a coated glass microscope slide. Phospho Explorer antibody array experiments and analyses were performed as a custom service by E-Biogen (Ebiogen Inc., Seoul, Republic of Korea).

Molecular Mechanisms of Diabetic Neuropathy: A Transcriptional Profling Study of db/db Mouse Sciatic Nerve

Project description:The mechanisms of diabetic neuropathy (DN) development and progression are not fully understood. We examined global gene expression in the sciatic nerve (SCN) of BKS-db/db mice at 8 and 24 weeks of age to identify genetic pathways altered in peripheral nerves at early and advanced stages of DN. The sets of differentially expressed genes were analyzed to identify enriched biological functions and regulated genetic pathways. Our results suggest that carbohydrate metabolism and lipid metabolism pathways are dysregulated in the db/db SCN at 8 weeks of age. Impairment of Schwann cell-extracellular matrix interaction and axon guidance occurs early in the development of DN, while neurotrophic support, neurotransmitter transport and axonogenesis are impaired at the advanced stage of DN. Gene expression changes also suggest that oxidative stress- and inflammation-mediated nerve damage occurs by 8 weeks. Our analysis identified mechanisms regulated in the early stages of DN. Additionally, interactions between genes from different pathways provide insight into potential relationships among the regulated pathways. RNA extracted from the SCN of four groups of mice: (8 week old db/db (n=8) or db/+ (n=8), and 24 week old db/db (n=6) or db/+ (n=7)) was hybridized to Affymetrix GeneChip microarrays. Gene expression for the two genotypes (db/db vs. db/+) was compared at each age. Gene expression at the early and advanced stages of DN (8 weeks vs. 24 weeks) was also compared for each genotype.

The eWAT proteomics of wild type mice and Mdm2 adipocyte-specific knock-in mice fed high-fat diet

Project description:The epididymal adipose tissue (eWAT) samples from wild type mice fed high-fat diet for 12 weeks (H_WT_E) and Mdm2 adipocyte-specific knock-in mice fed high-fat diet for 12 weeks (H_KI_E) were mixed separately, and subjected to proteomic study by Label-free quantitative techniques and mass spectrometry-based proteomics techniques, etc. The proteomics of mixed eWAT samples were performed in Jingjie PTM BioLab (Hangzhou) Co. Ltd (www.ptm-biolab.com.cn). The difference was determined by 1.5-fold-change criterion, FDR < 0.01.
