Self-reactivity of CD8 T-cell clones determines their differentiation status rather than their responsiveness in infections

ABSTRACT: The peripheral blood mononuclear cells (PBMCs) from young and old C57BL/6J mice were single cell RNA sequenced along with V(D)J sequences and cell surface protein markers that were used to demultiplex them from separate and unrelated experiment. The resulting data were then used to create an atlas of mouse peripheral CD8+ T cells. The analysis of this atlas has revealed two unconventional populations of antigen-inexperienced T cells and allowed for further examination of the steady-state phenotype of monoclonal T cells with various levels of self-reactivity.
