Expression of HCT116 cells transfected with CD44 siRNAs relative to that of HCT116 cells transfected with a control siRNA

ABSTRACT: To examine whether depletion of CD44 might affect antioxidant gene expression in cancer cells, HCT116 cells were transfected with CD44 or control siRNAs and subjected to cDNA microarray analysis. Although the CD44 siRNA was found to deplete the cells of CD44 mRNA, the expression of antioxidant genes was largely unaffected.

CD44+ or CD44– tumor cells isolated from K19-Wnt1/C2mE mice

Project description:We hypothesized that CD44+ tumor cells might have an increased capacity for reactive oxygen species (ROS) defense in the gastric tumors of K19-Wnt1/C2mE mice. To address this possibility, we examined the expression of antioxidant genes in tumor cells isolated from K19-Wnt1/C2mE mice by fluorescence-activated cell sorting (FACS). Lineage marker (Lin)–negative cells that were CD44+ or CD44– were thus isolated from the gastric tumors of 30-week-old K19-Wnt1/C2mE mice and subjected to cDNA microarray analysis. The expression of several key antioxidant genes, including those for glutathione peroxidase (GPX) and peroxiredoxin (PRDX) isoforms, was found to be increased in CD44+ tumor cells compared with that in CD44– cells.

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are using RNA sequencing analysis to evaluate the effects of si-LINC00152 on the mRNA of human colon cancer cell line HCT116. Methods: Human colon cancer cell line HCT116 was transfected with a control non-targeting siRNA to cells or transfected with siRNA targeting LINC00152 for 36 hours in DMEM medium (with 10% serum). Total RNA were extracted and detected by Illumina high-throughput RNA sequencing data analysis. 2 independent biological replicates were plated, transfected in parallel for each control and siRNA. Results: Log-fold changes of up- or down-regulated mRNAs between the control and experiment group were selected with a significance threshold of p<0.05. There are 90 mRNAs were up-regulated and 159 were down-regulated in “si-LINC00152” group comparing to “control” group. Conclusions: Our study describes the mRNA changes of human colon cancer cell line HCT116 transfected with LINC00152 siRNA.

Project description:Werner syndrome (WS) is a rare disorder characterized by the premature onset of a number of age-related diseases. The gene responsible for WS when mutated is believed to be involved in different aspects of transcription, replication, and/or DNA repair. We depleted the human WRN protein in HEK293 cells by transfecting them with small interference RNAs (siRNAs) against the WRN mRNA. The transcription profile of these transfected cells was compared to the transcription profile of HEK293 cells transfected with a scrambled siRNA that does not deplete the WRN protein.
